摘 要:目的:观察水杨酸钠致耳鸣大鼠模型海马区促肾上腺皮质激素释放因子1型受体(CRF1R)的表达。方法:24只大鼠随机分为3组,每组8只。A组:腹腔注射10%水杨酸钠溶液350mg/(kg·d),持续21d;B组:腹腔注射10%水杨酸钠溶液350mg/(kg·d),持续14d;C组:腹腔注射等量的生理盐水,持续14d。给药前2天、首次给药后2h和给药结束后对每只大鼠进行ABR测定,免疫组织化学方法以及Western Blot方法检测CRF1R在各组大鼠海马区的表达情况。结果:给药前2天各组大鼠ABR反应阈差异无统计学意义(P>0.05);首次给药后2hA组、B组大鼠反应阈分别较给药前提高了25.90dB SPL和25.03dB SPL(均P<0.01);给药结束后,A组、B组大鼠反应阈分别较给药前提高了34.91dB SPL和32.62dB SPL(均P<0.01),C组大鼠各时间点反应阈无明显变化(P>0.05)。免疫组织化学以及Western Blot方法显示大鼠脑海马区CRF1R的表达量A组>B组>C组(P<0.05)。结论:水杨酸钠致耳鸣大鼠脑海马区CRF1R的表达增加,且随注射时间的延长而增加,说明CRF1R可能在边缘系统参与耳鸣这一过程中发挥了重要作用。Objective:To observe the expression of corticotropin-releasing factor-1receptor in hippocampus of rats model of salicylate induced tinnitus.Method:Twenty-four rats were randomly divided into three groups,eight for each group.For Group A and Group B,10% salicylic sodium solution was intraperitoneal injected each day at the dose of 350mg/kg for 21 days in Group A and 14 days in Group B.Group C received intraperitoneal injection with the same amount of saline solution each day for 14 days.ABR were tested 2days before,and 2hours after the first administration and after the last injection.Immunohistochemical test and Western Blot were utilized to detect the expression of CRF1 Rin hippocampus for each group.Result:ABR thresholds tested 2days before the first administration of the 3groups showed no statistically significant difference(P〉0.05).At the time point of 2hours after the first injection,the ABR thresholds of Group A and Group B rose by 25.90 dB SPL and 25.03 dB SPL compared with that before the administration,respectively(P〈0.01).After the last administration,the ABR thresholds of Group A and Group B rose 34.91 dB SPL and 32.62 dB SPL compared with that before the administration,respectively(P〈0.01).The ABR thresholds of Group C showed no significant statistical difference at all the tested time points(P〉0.05).Immunohistochemical test and Western Blot revealed that the expression level of CRF1 Rin the hippocampus was A〉B〉C(P〈0.05).Conclusion:The expression of CRF1 Rin the hippocampus of salicylate induced tinnitus rat increased with the injection time,illustrating that CRF1 Rmay participate in the mechanism of tinnitus involving the limbic system.
