艾芬地尔对强迫游泳模型小鼠抗抑郁作用实验研究

摘　　要：目的：评价艾芬地尔对强迫游泳模型小鼠抗抑郁作用效果。方法：采用小鼠强迫游泳试验研究艾芬地尔抗抑郁作用。（1）艾芬地尔抗抑郁作用起效时间和持续时间试验：150昆明小鼠随机分为15组，每组10只，其中实验组（酒石酸艾芬地尔1mg·kg-1）、空白对照组（O．9％氯化钠注射液）、阳性对照组（氯胺酮2mg·kg-1），分别在腹腔注射给药后30min、60rain、120min、240min、300min进行强迫游泳试验；（2）艾芬地尔抗抑郁作用量效关系试验：100只昆明小鼠随机分为10组，每组10只，其中实验组（酒石酸艾芬地尔0．2mg·kg-1、1mg·kg-1、5mg·kg-1），阳性对照组（氯胺酮2mg·kg-1），空白对照组（0．9％氯化钠注射液），分别于腹腔注射后120min、240min进行强迫游泳试验；（3）反复给药后艾芬地尔抗抑郁效果评价：60只昆明小鼠随机分为6组，每组10只，其中实验组（酒石酸艾芬地尔5mg·kg-1），空白对照组（0．9％氯化钠注射液），第1、2、3组分别连续给药2d、4d、6d，三组均在最后一天给药后240min进行强迫游泳试验。结果：艾芬地尔可在120min内显著缩短强迫游泳小鼠累计不动时间，该作用可持续240min，其作用强度在1nag·kg-15mg·kg-1范围内与剂量呈正相关；反复多次给药后，其作用持续时间仍然为240min。结论：艾芬地尔对强迫游泳模型小鼠具有明显的抗抑郁作用，反复多次给药后，其抗抑郁作用持续时间维持稳定。Objective： To investigate the anti-depression effects of ifenprodil tartrate on the forced swimming mice model. Methods. Forced swimming test（FST） on mice was used to investigate the anti-depression effect of ifenprodil. （1） Test of the onset time and the duration time of ifenprodil tartrate in the antidepressant effect, one hundred and fifty mouse were randomly divided into fifteen groups（n= 10）, including the experimental groups（1 rng · kg-1 ifenprodil tartrate）, the positive control groups（2 mg · kg-1 ketamine） and the control groups（NS）. The forced swimming test were taken after 30 minutes, 60 minutes, 120 minutes, 240 mi- nutes and 300 minutes respectively and the immobility time of mouse were recorded. （2） Test of the dose-effect relationship of ifen- prodil tartrate in the antidepressant effect：One hundred mouse were divided randomly into ten groups（n~ 10）, including the experi- mental groups（0. 2 mg · kg-1 , 1 mg · kg-1 and 5 mg · kg-1 ifenprodil tartrate）, the positive control groups（2 mg · kg-1 ketamine） and the control groups（NS）. The forced swimming test was taken after 120 minutes and 240 minutes respectively. （3） Test of the an- tidepressant effect evaluation of ifenprodil tartrate on repeated dose： sixty mouse were randomly divided into six groups（n= 10）, in- cluding the experimental groups（5 mg ~ kg-1 ifenprodil tartrate） and the control groups（NS）. The experimental group mouse were intraperitoneal injectioned ifenprodil tartrate for two days, four days and six days, respectively. The forced swimming tests were taken after 240 minutes at the last day of injection. Results： The onset time and the duration time of the effect of anti-depression of if- enprodll were respectively 120 minutes and 240 minutes. There was a positive correlation between the dose-dependent relationship and the antidepressant effect of ifenprodil tartrate in the range of 1 mg · kg-1 ---5 mg · kg-1.
