摘  要:目的了解急性肺损伤(ALI)小鼠肺组织中水通道蛋白5(AQP5)的表达变化,探讨半夏提取物(EP)治疗ALI的药理机制,为半夏的临床应用提供理论基础。方法 36只昆明种小鼠随机分成3组,即空白对照组(NS组)、肺损伤造模组(ALI组)和半夏提取物治疗组(EP组),6 h后观察测定肺组织湿干重比值(W/D),HE染色光镜下观察肺炎症变化,ELISA法检测小鼠血清TNF-α浓度,RT-PCR检测肺组织匀浆AQP5 mRNA表达情况。结果 ALI组小鼠肺部炎症反应显著,中性粒细胞浸润为主,同时肺泡明显充血水肿。EP组肺组织炎症减轻、充血消退。与NS组和EP组比较,ALI组的W/D值差异有统计学意义(P<0.05);ALI组肺组织AQP5 mRNA的表达,较NS组显著降低,较EP组的表达显著升高(P<0.05)。ALI组血清TNF-α的表达,较NS组显著升高,较EP组的表达显著降低(P<0.05)。结论半夏提取物可通过上调肺AQP5的表达减轻肺水肿,作用机制可能是通过抑制炎症因子TNF-α的表达实现的。Objective To investigate lung tissue aquaporin 5( AQP5) expression changes in acute lung injury( ALI)mice,and to explore the pharmacological mechanism of extracts of pinellia( EP) in the treatment of acute lung injury.Methods 36 Kunming mice were randomly divided into three groups,namely the blank control group( NS group),lung injury model group( ALI group) and pinellia extract treatment group( EP group). After six hours,lung tissue wet / dry weight ratio( W / D) were observed and determined,lung tissue histopathological changes were observed by HE staining,serum concentrations of TNF-α in mice were detected by ELISA and lung tissue AQP5 mRNA expressions were detected by RT-PCR. Results Reflection of lung inflammation in mice in ALI group was significant,with main neutrophil infiltration as well as marked alveolar edema and hyperemia. Lung inflammation in EP group was alleviated and the edema was reduced. Compared with the NS group and EP group,W / D value difference was statistically significant in ALI group( P〈 0. 05); Compared with NS group,AQP5 mRNA expression decreased significantly and TNF-αin blood were higher in ALI group( P〈 0. 05). Compared with ALI group,AQP5 mRNA expression increased significantly and TNF-α in blood were lower in EP group( P〈 0. 05). Conclusion EP can reduce pulmonary edema by upregulating the expression of lung AQP5 which whose mechanism may be related with the inhibition of TNF-α.
