摘 要:目的制备胶原-纤维蛋白胶双层复合材料,评估其作为皮肤组织工程材料复合人脐带间充质干细胞(h UCMSC)修复小鼠全层皮肤创伤的效果。方法制备胶原-纤维蛋白双层材料,扫描电镜检测双层支架材料表面结构。24只小鼠建立全层皮肤创面模型后,随机分为3组:空白组用无菌纱布处理伤口、h UCMSC复合胶原组和h UCMSC复合胶原-纤维蛋白胶组,分别采用各组敷料处理创面。治疗后5、10、15 d,观察创面愈合情况,计算3组创面愈合率并进行对比。治疗后15 d,取小鼠创面新生皮肤组织苏木精-伊红染色,观察新生皮肤组织结构。结果扫描电镜结果显示:双层材料胶原层横断面呈现出高孔径结构并且互相连通,纤维蛋白胶层横断面显示紧密的三维网状结构;创面愈合结果显示:h UCMSC复合胶原-纤维蛋白胶组治疗5、10、15 d创面愈合率分别达到(62.62±6.27)%、(88.47±7.14)%、(96.14±4.56)%,优于空白组(t=17.03、16.22、15.69,P均<0.01),同时也优于h UCMSC复合胶原组(t=2.71、5.60、5.28,P均<0.05)。h UCMSC复合胶原-纤维蛋白胶组组织学检查新生皮肤组织结构良好。结论胶原-纤维蛋白胶双层支架具有较好的三维空间结构,复合h UCMSC后有利于促进皮肤创面愈合。Objective Preparation of collagen- fibrin double layer membrane. To study the effect of human umbilical cord mesenchymal stem cell (hUCMSC) combined with collagen-fibrin double layer membrane on wound healing in mice. Methods Preparation of collagen-fibrin double layer membrane. The surface stueture of the double loyer membrane was investigated by SEM. Twenty-four mouse were prepaird as full thickness skin wound model and randomly divided into 3 groups: control group was bound up their wounds by conventional method, hUCMSC combined with collagen membrane treated group and hUCMSC combined with collagen-fibrin double layer membrane treated group. Results The SEM results showed that collagen-layer exhibited a highly porous and interconnected structure. The wound healing rate in double layer membrane combined with hUCMSC treated group at 5,10,15 day was (62.62±6.27 ) % , ( 88.47±7.14)% ,(96.14±4.56)%, better than control group (t = 17. 03, 16. 22, 15.69, P 〈 0.01), and hUCMSC combined with collagen membrane treated group (t = 2. 71, 5.60, 5.28, P 〈 0. 05 ), differences were significant. Conclusions Collagen-fibrin double layer membrane showed a good three dimensional space structure, combined with hUCMSC could accelerate wound healing.
