子宫内膜癌患者癌组织中PCNA和CD_(34)蛋白表达的实验研究

摘　　要：目的:探讨子宫内膜癌患者癌组织中PCNA和CD34蛋白的表达及意义。方法:选取子宫内膜癌手术患者120例,术后取手术切除的癌组织(实验组)及癌旁正常组织(对照组),采用ELISA法和实时荧光定量PCR法检测患者癌组织中PCNA和CD34蛋白的表达。结果:与对照组比较,实验组患者癌组织中PCNA、CD34的蛋白含量及m RNA水平均明显升高,差异有统计学意义(P<0.01);PCNA和CD34在组织中蛋白含量及m RNA水平均呈正相关性(r=0.608、P=0.037,r=0.576、P=0.029)。结论:子宫内膜癌患者癌组织中PCNA和CD34蛋白表达的升高,可能与子宫内膜癌的发生、发展有关,其作用机制有待于进一步研究。Objective： To explore the expressions and significance of PCNA and CD34 proteins in cancer tissue of patients with endometrial cancer. Methods： A total of 120 patients with endometrial cancer were selected,the resected cancer tissue specimens were designed as experimental group,while the resected normal tissue specimens adjacent to cancer were designed as control group; ELISA and real-time fluorescence quantitative PCR were used to detect the expressions of PCNA and CD34 proteins. Results： Compared with control group,the levels of PCNA and CD34 proteins and m RNA in experimental group increased significantly,there were statistically significant differences（ P〈0. 01）; PCNA proteins and m RNA levels were positively correlated with CD34 proteins and m RNA levels（ r = 0. 608,P =0. 037; r = 0. 576,P = 0. 029）. Conclusion： The high expressions of PCNA and CD34 proteins in cancer tissue of patients with endometrial cancer may be related to occurrence and development of endometrial cancer,the mechanism needs further study.
