摘 要:目的检测Graves病人外周血细胞因子和树突状细胞(DCs)不同亚群的数量和表达情况,初步探讨DCs细胞亚群在疾病中的作用。方法利用流式细胞仪检测30例Graves病患者和30名健康体检者的外周血树突状细胞亚群数量,同时利用ELISA检测肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)、白细胞介素-2(IL-2)及IL-6等细胞因子的水平。结果测得Graves组髓样树突状细胞(p DC)和浆细胞样树突状细胞(m DC)值均高于正常对照组。ELISA检测细胞因子发现,Graves患者血清中TNF-α、IFN-γ、IL-6水平均高于正常对照组,其中TNF-α显著升高(P<0.01),而IL-2水平显著低于对照组(P<0.01)。结论 Graves病人中p DC和m DC水平升高,细胞因子分泌失调,Th1/Th2失衡,从而导致机体自身免疫反应的产生。Objective To discuss the role of DC cells in Graves disease,we detect the frequency of different DC subsets and the secretion of cytokines in Graves patients' peripheral blood. Method Flow cytometry( FCM) was performed to assess the frequency of different DC subsets in peripheral blood of 30 Graves patients and 30 healthy cases respectively. And ELISA was applied to detect the secretion of TNF-α,IFN- γ,IL-2 and IL-6 and other cytokines in the peripheral blood. Results The frequency of p DCs and m DCs in Graves disease group was higher than that in healthy cases.And ELISA results showed that the secret of TNF-α,IFN- γ and IL-6 in Graves disease patients' peripheral blood were significant higher than that in normal control group,while IL-2 were significantly lower than the control group( P〈0. 01).Conclusion The frequency of p DCs and m DCs in Graves disease were higher than that of healthy controls,which indicated that p DC and m DC were involved in the pathogenesis of Graves.
