天丝饮的抗抑郁作用及其对IDO的调节

摘　　要：目的研究天丝饮的抗抑郁作用及其对色氨酸(TRP)、犬尿氨酸(KYN)途径关键代谢酶吲哚胺2,3过氧化酶(IDO)的影响。方法使用脂多糖(LPS)腹腔注射1 mg/kg造成小鼠抑郁模型,采用Noldus Etho Vision XT9系统采集分析小鼠强迫游泳不动时间,采用HPLC-MS/MS技术检测脑组织色氨酸、犬尿氨酸含量,实时荧光定量PCR检测脑组织中IDO、NF-κB的mRNA表达水平。结果 LPS注射4 h后可以引起模型组小鼠不动时间延长(P<0.05),脑组织IDO活性增高,IDO和NF-κB mRNA表达增多(P<0.01)。天丝饮能缩短小鼠不动时间(P<0.05),抑制IDO活性及IDO、NF-KB mRNA表达数量(P<0.01)。结论天丝饮具有抗抑郁药物样作用,这一作用与抑制炎症因子诱导的IDO激活有关,天丝饮通过抑制NF-κB对IDO的直接和间接诱导作用而调节了IDO的活性和表达。Objective To study the effect of antidepression of Tiansi Liquid, a TCM basic prescription for treating amnesia and palpipation, which influences on indoleamine 2, 3-dioxygenase （IDO）, the key metabolism enzyme of the tryptophan-kynurenine pathway. Methods The mice model of depression was established by intraperitoneal injection of 1 mg/kg lipopolysaccharide （LPS）. The immobility time in forced swimming test was collected by Noldus EthoVision xrIx） system. The content of tryptophan and kynurenine in brain tissue were detected by HPLC-MS/MS technique. The mRNA expression level of iDO and NF-KB in brain tissue were measured by using real-time PCR. Results Four hours after the injection of LPS in model group, the immobility time of mice was prolonged （ P 〈 0.05 ） , while the activ- ity of IDO in brain tissue and the mRNA expression level of IDO and NF-κB increased significantly（P 〈 0.01 ）. Tiansi liquid shortened the immobility time of mice （ P 〈 0.05 ） , and inhibited the activity of IDO and the expression of IDO and NF-KB mRNA （P 〈0.01）. Conclusion Antidepressant-like drug effect of Tiansi liquid is due to inhabiting directly and indirectly effects of NF-κB on IDO which is activa- ted by inflammatory cytokines.
