大黄酚固体分散体对大鼠酒精性肝损伤的保护作用研究

摘  要:目的研究大黄酚固体分散体对酒精肝性损伤大鼠的保护作用。方法健康SD大鼠48只,随机分为空白对照组、模型组、大黄酚给药组、大黄酚固体分散体给药组,除空白对照组外,各组大鼠均灌胃高度白酒,各给药组大鼠灌胃给予大黄酚、大黄酚固体分散体。给药8周后,取动物血清及肝脏组织,测定丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)的活力,测定丙二醛(MDA)含量。结果与大黄酚组比较,大黄酚固体分散组大鼠体内谷草氨酸氨基转移酶(AST)显著降低、谷丙氨酸氨基转移酶(ALT)、超氧化物歧化酶(SOD)活性显著升高,肝脏中丙二醛(MDA)含量下降。结论以聚乙二醇6000为载体制备的大黄酚固体分散体,对酒精性肝损伤大鼠有明显的保护作用。Objectives To study the protective effects of chrysophanol solid dispersions on alcoholic liver in jury in rats. Methods Forty - eight healthy SD rats were randomly divided into normol group, model group, chrysophanol group, chrysophenol solid dispersions group. In addition to the control group, each group of rat was fed with alcohol'and chrysophanol, chrysophanol solid dispersions. After 8 week, the alanine aminotransferase (ALT), aspartate aminotransferase (AST), superoxide dismutase (SOD) activity and content of malondialdehyde (MDA) in serum and liver were determined. Results Compared with chrysophanol group, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity, reduce malondialdehyde (MDA) content were significantly reduced in chrysophanol solid dispersion group, but their activity of super oxide dismutase (SOD) was increased. Conclusion The chrysophanol solid dispersions prepared with polyethylene glycol 6000 as the carrier has an obvious protective effect on alcoholic liver injury in rats.
