摘  要:目的探讨丹参多酚对铁过载引起的脐血造血细胞氧化应激损伤的保护作用。方法分离脐血单个核细胞,分为对照组、模型组、丹参多酚低、中、高剂量组。模型组及丹参多酚低、中、高剂量组以400μmol/L浓度的枸橼酸铁胺(FAC)建立铁过载细胞模型,丹参多酚低、中、高剂量组于培养第3天分别加入18、50、160μg/m L丹参多酚培养24 h;对照组不加FAC及丹参多酚。采用流式细胞术检测各组细胞的活性氧(ROS)水平、细胞凋亡率,计数各组细胞集落形成数量。结果模型组ROS水平高于对照组及丹参多酚各剂量组(P均<0.05)。模型组细胞凋亡率高于对照组及丹参多酚高剂量组(P<0.05)。模型组红系祖细胞集落形成单位(CFU-E)、粒—单核系祖细胞集落形成单位(CFU-GM)、爆式红系祖细胞集落形成单位(BFU-E)和混合祖细胞集落形成单位(CFU-mix)数量均低于对照组及丹参多酚高剂量组(P均<0.05)。结论丹参多酚可减轻氧化应激引起的细胞凋亡及其对造血细胞增殖、分化功能的抑制作用。Objective To investigate the protective effect of salvianolate on the iron overload-derived oxidative stress injury of mononuclear cells of umbilical cord blood (UC-MNCs).Methods The UC-MNCs were divided into the control group, FAC group, low-dose salvianolate group, medium-dose salvianolate group, and high-dose salvianolate group.The control group received normal culture .The iron overload cell models in the FAC and salvianolate groups were established by adding 400 μmol/L FAC into the culture medium for 24 h on the second day.Then the low-dose, medium-dose and high-dose salvianolate groups were received 18, 50 and 160μg/mL salvianolate for 24 h, respectively on the third day .Then the levels of reactive oxygen species ( ROS) , apoptosis rate and LIP level were detected by flow cytometry , respectively .Re-sults The ROS level of the FAC group was higher than that of the control , the low-dose, medium-dose and high-dose salvianolate groups (all P〈0.05).The apoptosis rate in the FAC group was higher than that of the control group and high-dose salvianolate group(P〈0.05).The counts of colony-forming unit-erythroid (CFU-E), granulocyte-macrophage colo-ny-forming units ( CFU-GM) , erythropoietic burst-forming units ( BFU-E) and mixed progenitor cell colony-forming units ( CFU-mix) in the FAC group were significantly lower than those of the control group and high-dose salvianolate group ( P〈0.05).Conclusions Salvianolate can reduce the oxidative stress caused by apoptosis and inhibition of proliferation and differentiation of hematopoietic cells .
