摘 要:目的分析大连市引起儿童手足口病(hand-foot-mouth Disease,HFMD)的肠道病毒71型(Enterovirus71,EV71)VP1基因特征,寻找与毒力相关的基因位点。方法用逆转录-聚合酶链反应(RT-PCR)方法对HFMD轻症和重症临床标本进行EV71的VP1全基因扩增,分析扩增产物同源性,并与各基因型代表株构建进化树,比对分析轻症和重症间EV71的VP1基因。结果扩增产物核苷酸和蛋白质序列与C4a亚型具有较高的同源性,在系统进化树上,与C4a亚型处于同一分支。轻症和重症EV71的VP1基因无共同差异位点。结论大连市HFMD病原体EV71为C4a亚型,EV71存在多种传播链,在VP1基因上未发现与毒力相关的基因位点。Objective To analyze the genotype and virulence site of Enterovirus 71 from children with hand-foot- mouth disease (HFMD) in Dalian City. Methods The complete VP1 gene of EV71 was amplified from specimens of the mild and severe patients with HFMD in Dalian by reverse transcription-polymerase chain reaction (RT-PCR), sequenced and analyzed for homology. The phylogenetic tree was constructed with genotypes based on nucleotide sequences of the VP1 gene. The sequence of complete VP1 gene was compared between mild with severe patients with HFMD. Results There was a high homology in nucleotide and protein sequence between the amplified products and C4a subgenotype. The amplified sequences were clustered with C4a subgenotype in the phylogenetic tree. Conclusion The EV71 caused HFMD in Dalian belonges to subgenotype C4a. There are several transfer-chains and no determinants of virulence in VP1 gene of EVTI in Dalian.
