不同处理条件下绞股蓝多糖含量的测定与比较

摘　　要：(目的)通过比较绞股蓝饮片、绞股蓝超微粉以及绞股蓝发酵液中绞股蓝多糖的含量,为绞股蓝的临床应用提供依据。(方法)分别采用水煎煮法,水提法,醇碱提法、醇处理水提法和石油醚醇处理水提法5种不同方法,对绞股蓝饮片中的绞股蓝多糖含量进行了测定及比较,确定最佳提取方法。采用所确定的最佳处理方法测定绞股蓝超微粉中绞股蓝多糖含量。而绞股蓝发酵液直接采用醇沉处理测定其中的多糖含量,并对结果进行分析比较。(结果)五种提取方法以醇碱提法提取绞股蓝粗多糖含量最高为49.1400mg/g,纯度最高为0.2068g/g;绞股蓝叶超微粉提取的绞股蓝粗多糖含量为158.5500mg/g,纯度为0.2448g/g;二次发酵液提取的绞股蓝粗多糖含量为1638.5500mg/g,纯度为0.6161g/g。(结论)五种提取方法中以醇碱提法提取的绞股蓝多糖含量和纯度最高;绞股蓝超微粉中的绞股蓝多糖含量明显高于绞股蓝饮片中的,但经过发酵处理的绞股蓝中的多糖含量最高。（Purpose）According to compare polysaccharide content of Gynostemma pentaphylla from decocting pieces,ultrafine powders and fermentation broth, then apply the bases for clinical application of Gynostemma pentaphylla.（Methods）Respectively use five extracted methods namely water decocting method,water-extracted method,alcohol-alkali extracted method,alcohol disposed-water extracted method and aether petrolei- alcohol disposed,water-extracted method to compare polysaccharide content of Gynostemma pentaphylla, then confirm the best method.Use the best method to extract polysaccharide form Gynostemma pentaphylla ultrafine powders and directly use alcohol precipitation to extract the polysaccharide in fermentation broth of Gynostemma pentaphylla,then compare the content of them.（Consequence） The content of polysaccharide extracted by alcohol-alkali extracted method is 49.1400 mg/g and the purity is 0.2068 g/g,which is the most and highest in the five methods.The content of polysaccharide in Gynostemma pentaphylla ultrafine powders of leave is 158.5500 mg/g and the purity is 0.2448 g/g. The content of polysaccharide in fermentation broth of Gynostemma pentaphylla is 1638.5500mg/g and the purity is 0.6161 g/g.（Conclusions） Content and purity of polysaccharide extracted by alcohol-alkali extracted method is the best.Polysaccharide content of Gynostemma pentaphylla ultrafine powders is more than decocting pieces＇ s and polysaccharide content of fermentation broth is the maximum.
