糖毒清颗粒对糖尿病肾病肾功能不全大鼠肾组织TGF-β1、Smad3及Smad4 mRNA表达的影响

摘  要:目的研究糖毒清颗粒对糖尿病肾病肾功能不全(Diabetic renal insufficiency,DRI)大鼠肾组织TGF-β1、Smad3及Smad4 mRNA表达的影响。方法32只Wister大鼠随机分为3组:正常对照组、模型组及糖毒清组。除正常对照组外,采用链脲佐菌素+腺嘌呤灌胃制作DRI大鼠模型,糖毒清组予糖毒清颗粒(4.32g·kg-1·d-1)灌胃,模型组和正常对照组均予等量生理盐水灌胃,连续2个月。检测肾功能中尿素氮(BUN)、血肌酐(Scr)等指标及肾脏组织学观察,并采用RT-PCR法检测肾组织TGF-β1、Smad3及Smad4的mRNA表达。结果糖毒清颗粒组TGF-β1、Smad3、Smad4的mRNA表达明显低于模型组(P<0.05),与正常对照组比较,差异无统计学意义(P>0.05)。结论糖毒清颗粒可明显降低DRI大鼠肾组织中TGF-β1、Smad3、Smad4的mRNA表达,这可能是糖毒清治疗DRI、保护肾功能的分子机制之一。Objective To explore the effect of Tangduqing granules(TG) on the expression of TGF-β 1,Smad3 and Smad4 mRNA in kidney tissue of diabetic renal insufficiency(DRI) rats.Methods Thirty-two Wistar rats were randomized into normal control group,DRI model group and TG group.DRI rat model was induced by intragastric gavage of streptozotocin(STZ) and adenine.Both normal control group and DRI model group were given intragastric administration with equal volume of normal saline,and TG group was given intragastric administration with TG 4.32 g/kg·d-1.Two months later,renal function was detected,the pathological changes of kidney tissue were examined,and the renal expression of TGF-β 1,Smad3 and Smad4 mRNA was detected by RT-PCR.Results Compared with DRI model group,the expression of TGF-β 1,Smad3,Smad4 mRNA of TG group was significantly decreased(P〈0.05),but the expression level was similar to that of the normal control group(P〉0.05).Conclusion Tangduqing granules can obviously reduce the expression of TGF-β 1,Smad3 and Smad4 mRNA in kidney tissue of DRI rats,which may be the therapeutic and renal protective mechanism of Tangduqing granules in DRI rats.
