摘 要:目的以常规药理学指标研究败酱草总皂苷(BJC)对溃疡性结肠炎的药效,同时考察其尿液代谢表型差异。方法 SD大鼠随机分为BJC组、模型组和正常组,检测相关生化指标,尿样采用超高效液相色谱-四级杆飞行时间质谱(UPLC/Q-TOf MS)检测分析,采用主成分分析(PCA)以及偏最小二乘判别法(PLSDA)对数据进行处理。结果与模型组比较,BJC组大鼠结肠组织学损伤明显减轻,超氧化物歧化酶(SOD)活性显著升高(P<0.01),而髓过氧化物酶(MPO)活性、丙二醛(MDA)水平显著降低(P<0.01)。代谢组学分析各组间区分明显,尤以PLSDA方法分组更加明显。结论代谢组学初步研究显示,败酱草总皂苷可明显改善大鼠溃疡性结肠炎,PLSDA数据处理方法可靠。Objective With the routine pharmacological parameters as the comparative indexes,the urine metabonomic profile were investigated to study the effect of Patrinia saponins(PC) on ulcerative colitis(UC).Methods The rats were randomly divided into PC group,trinitro-benzene-sulfonic acid(TNBS) group and black control group.The biochemical indicators of UC were determined after administration.The rat urine was analyzed by ultra performance liquid chromatography/quadrupole time-of-light mass spectrometry(UPLC/Q-TOf MS) and then subjected to data process with principal component analysis(PCA) and partial least square discrimination analysis(PLSDA).Results Compared with the model group,the colonic histological damage in PC group was much relieved,superoxide dismutase(SOD) activity was increased(P〈0.01),and myeloperoxidase(MPO) activity and malondialdehyde(MDA) level were decreased(P〈0.01).The metabonomics profile differences were obvious in different groups,and the results of PLSDA presented the differences more obviously.Conclusion The results of preliminarily metabonomic study indicates that Patrinia saponins show therapeutic effect on TNBS-induced colitis in rats,and PLSDA is a feasible method for data process.
