摘  要:以一株北里链霉菌(Streptomyces kitasatoensis)Zn1201为出发菌株,使用氯化锂前处理后照射紫外线进行复合诱变,通过L9(34)正交实验等处理结果得知,1.5%的氯化锂浸泡2 h、UV照射90 s得到的正突变株摇瓶效价提高较大,通过耐高浓度豆油和耐自身代谢物的选择性筛选,获得3株高产突变株,其中Zn1401突变株摇瓶发酵效价比出发菌株提高了14.3%,且该菌株传至5代,遗传性状稳定。该菌株应用于生产之后,产量提高7%左右,年新增利润1091万元。The Streptomyces kitasatoensis Znl201 is used as an original strain. Compound Mutation happens on it after LiCI soaking and uvio|izing. According to the result of L9(34) orthogonal experiment, we learn the assay of positive mutant strain increases a lot, which is caused by 1.5 % LiCI soaking for 2 hours and uviolizing for 90 seconds. Three high-yield mutant strains of tolerant high-concentration soya-bean oil and self-metabolites are obtained by selective screen. One of them is Znl401 mutant strain, which shake-flask culture assay increases by 14.3 % than the original strain and passes 5 generations under stable inheritable character The output of Kitasamcyin increases by 7 % and the profit increases by RMB 10,910,000.00 after using new strain into production.
