摘 要:目的探讨右旋美托咪啶与在体小鼠肺泡液体清除率(AFC)之间的关系,以明确其在肺脏液体清除中的作用。方法应用酶标仪测定伊文思蓝标记的小牛血清白蛋白浓度的方法测定小鼠在体AFC。结果与1mmol/L阿米洛利抑制效应相反,气管内注入1.5μg/kg右旋美托咪啶后,能显著增加小鼠在体AFC。与阿米洛利合用后此增强效应受到抑制,表明右旋美托咪啶能够增加与上皮钠通道有关的阿米洛利敏感性AFC。结论临床上对合并肺脏损害的患者进行镇静时,可以考虑右旋美托咪啶对肺脏液体清除作用的影响,应用其进行相应的治疗。Objective To explore the relationship between dexmedetomidine and alveolar fluid clearance (AFC) in mice in "oivo , and to determine its effects on the process of lung fluid clearance. Methods In vivo AFC was measured using Evans blue-labeled bovine serum albumin protein assays. Results Contrary to the obviously inhibitory effects of 1 mmol/L amiloride (a specific ENaC blocker), the rate of in vivo AFC was significantly in- creased in mice intratracheally administrated 1.5 μg/kg dexmedetomidine. In the presence of both dexmedetomi- dine and amiloride, the increase of fluid resolution was further inhibited, suggesting that dexmedetomidine could increase amiloride-sensitive fraction of AFC associated with ENaC. Conclusion These data demonstrate that the increase of lung fluid clearance following application of dexmedetomidine should be considered clinically when the patient is complicated with lung injury, and dexmedetomidine could be used in the therapy.
