摘  要:目的探讨亚甲蓝光化学法在冷沉淀病毒灭活中应用的可行性。方法将12袋新鲜血浆平均分为实验组及对照组,实验组加入终浓度1μmol/L亚甲蓝溶液按照50 mA,15 min进行病毒灭活处理,对照组加入等体积生理盐水不做灭活处理,分别按照常规方法制备冷沉淀,贮存7 d后分别检测亚甲蓝光化学法病毒灭活前后冷沉淀中凝血因子Ⅷ(clotting factorⅧ,FⅧ)和纤维蛋白原(fibrinogen,Fib)的含量。结果实验组FⅧ和Fib的含量分别为(193.79±24.95)IU/U,(85.00±6.50)mg/U,对照组FⅧ和Fib的含量分别为(218.55±16.61)IU/U,(114.00±9.70)mg/U,两组之间FⅧ含量无统计学差异,Fib含量具有统计学差异(P<0.05)。结论亚甲蓝光化学法病毒灭活方法是否可应用与冷沉淀病毒灭活,仍有待进一步考证。Objective To explore the use of methylene blue photochemical method in the virus inactivation of cryo precipitate.Methods Twelve packages of fresh plasma were averagely divided into experimental group and control group. Methylene blue solution with final concentration 1 μmol/L was added into the plasma and then the plasma was disposed by 50 mA light treatment for 15min in the experimental group.The same volume of saline was add into the control group without illumination inactivatioruBoth groups were prepared into the cryoprecipitate by conventional method.And the lev els of FⅧ and Fib in cryopreeipitate were detected respectively after storage for 7 days.Results The levels of FⅧ and Fib in the experimental group were (193.79 ± 24.95) IU/U and (85.00± 6.50) mg/U,respeetively and (218.55±16.61 ) IU/U and (114.00± 9.70) mg/U respectively in the control group. Fib and FⅧ was not significantly different between two groups.Conclusion Whether the methylene blue photochemical method can be used in the virus inactivation of cryopreeip- itate,still needs further research.
