口蹄疫病毒样颗粒作为抗癌药物载体的条件探索

摘　　要：[目的]探讨口蹄疫病毒样颗粒作为抗癌药物载体运载药物进入细胞的可行性。[方法]利用原核表达系统表达纯化口蹄疫结构蛋白,在组装缓冲液中形成口蹄疫病毒样颗粒,利用化学偶联试剂将抗癌药物偶联到病毒样颗粒的表面,形成的载体药物复合物处理细胞,荧光染色观察复合物进入细胞情况。[结果]口蹄疫病毒样颗粒作为新型抗癌药物载体,能够有效携带抗癌药物,该研究为设计新型药物载体提供了可能。[结论]口蹄疫病毒样颗粒具有作为新型药物载体的潜力。[ Objective ] To explore the formation of a new anticancer drug carrier in wtro as weir as to research its antjcancer ettect.[Method] Using prokaryotic expression system to express and purified FMDV structural proteins of VP0, VP3 and VPI, then self-assamble to form the FMDV virus-like particles in the assembly buffer, after that, coupling chemical anticaneer drug to the virus-like particle surface, forming a new anti-cancer drugs carrier. [ Result ] FMDV virus-like particles as a novel anti-cancer drug carrier is capable of porting anti-cancer drugs, then may providing a new approach for designing novel drug carrier. [ Conclusion] FMDV virus-like particles have potential as a novel drug carrier.
