摘  要:目的:探讨七味白术散及七味白术散提取物对肠道菌群失调小鼠小肠黏膜干扰素-α(IFN-α),白细胞介素-4(IL-4),白细胞介素-10(IL-10)表达的影响。方法:取ICR小鼠,采用头孢曲松钠和林可霉素联合中药番泻叶药液ig造模,观察小鼠大便性状、腹泻次数和粪便细菌培养结果确立造模成功后,将小鼠分成4组分别为模型组,七味白术散组(5.2 g·kg-1),七味白术散提取物组(0.94 g·kg-1),并另设正常组。各治疗组给予相应药物0.4 m L/只,每天ig 1次,正常组、模型组小鼠给予等量的蒸馏水ig。分别于造模的第3,7,9,11,13天心脏采血处死小鼠,取小肠行病理切片,采用免疫组织化学法观测小肠黏膜上皮细胞IFN-α,IL-4,IL-10的阳性表达。结果:与正常组比较,模型组IFN-α在3,7 d时表达增高(P<0.05),9,11,13 d表达逐渐下降,但仍然高于正常和各治疗组(P<0.05);七味白术散组与提取物组IFN-α的表达两组之间比较无统计学意义,但两组在各个时间点IFN-α均低于模型组(P<0.05);小鼠小肠黏膜IL-4,IL-10表达模型组明显低于正常组、低于各治疗组(P<0.05),七味白术散组和提取物组小鼠小肠黏膜IL-4,IL-10表达在第9天时明显增高,13 d时与正常组比较,差别无统计学意义,提取物组与七味白术散组比较差别无统计学意义。结论:七味白术散及其提取物均有降低肠道菌群失调小鼠小肠黏膜IFN-α,提高IL-4,IL-10的表达,从而抑制肠道炎症反应,减少免疫损伤作用。Objective: To observe the effect of Qiwei Baizhu San and its extract on the expressions of interferon-α(IFN-α), interleukin-4 (IL-4) and interleukin-10 (IL-10) in the small intestinal epithelial cell of enteric dysfunctions in mice. Method: The enteric dysfunctions model was induced in ICR mice by intragastric administration of ceftriaxone sodium, lincomycin and folium sennae decoction. The stool, diarrhea and fecal bacterial were observed. The mice were randomly divided into four groups: the model group (MG) , the Qiwei Baizhu San group (QG, 5.2 g·kg^-1), the Qiwei Baizhu San extract group (QTG 0. 94 g ·kg^-1), and the normal control group ( NG). The mice in the treatment groups received 0.4 mL volume of the corresponding medicines and the mice in the MG, NG received the same volume of distilled water. The mice were sacrificed at 3, 7, 9, 11, 13 days after modeling. The positive expressions of IFN-α, IL-4 and IL-10 in small intestinal epithelial cell were observed by using immunohistochemistry assay. Result: Compared with the NG, the expression of IFN-α increased at day 3 and 7 (P 〈 0.05 ) , then gradually decreased at day 9, 11 13, while the results were still higher than the NG, QG and QTG (P 〈0.05). The levels of IL-4, IL-10 decreased in the NG, QG and QTG (P 〈0.05).Compared with MG, at day9 (P〈0.05) Baizhu San and its expression of IFN-α, the expression of IFN-α decreased at all days (P 〈 0. 05) , the levels of IL-4, IL-10 increased , and these results had no significant difference between QG and QTG. Conclusion: Qiwei extracts could inhibit the intestinal inflammation and reduce immune injury by reducing increasing the levels of IL-4, IL-10 in the small intestine.
