摘 要:采用超高效液相色谱法(UPLC),以甲醇-0.1%磷酸水溶液梯度洗脱,对山西、河南两省共30批香加皮中的3种成分(绿原酸、4-甲氧基水杨醛、杠柳毒苷)进行同时测定,并对其指纹图谱进行研究。结果表明不同产地样品中,3种成分含量差异较大;香加皮药材指纹图谱共标定17个共有峰,除H3,H11,样品相似度均在0.90以上。以3种成分含量及相似度数据进行显著性差异分析、聚类分析及主成分分析,结果表明不同产地的香加皮药材质量存在一定差异。建立的含量测定结合指纹图谱分析方法可更好地用于香加皮的质量评价。Ultra performance liquid chromatography (UPLC) was employed for simultaneous determination of three components and fingerprint analysis of Periplocae Cortex with gradient elution of mehtanol and water containing 0. 1% phosphoric acid as mobile phase. Three components including ehlorogenic acid, 4-methoxysalieylaldehyde and periplocoside were well separated under the analytical con- dition. Seventeen peaks were selected as the common peaks of 30 batches of Periplocae Cortex. The results showed that there is a sig- nificant difference in contents of periplocoside between the samples collected from Henan and Shanxi province. Based on the results of three components quantification and fingerprint analysis, hierarchical clustering analysis (HCA) and principle component analysis (PCA) were used to further prove the differences between two group samples, and the results indicated that quality of Periplocae Cortex from Shanxi was more stable than that from Henan. The established UPLC fingerprint and quantitative analysis methods could be used efficiently in the quality control of Periplocae Cortex, and this study might contribute to the reasonable clinical application.
