HPLC-ELSD法测定阿胶三宝膏中黄芪甲苷的含量

摘　　要：目的：建立测定阿胶三宝膏中黄芪甲苷含量的高效液相色谱联合蒸发光散射检测方法 。方法：采用FORTIS-C18柱（4.6 mm×250 mm,5μm）,以乙腈：0.05%磷酸二氢钾溶液（21∶79）为流动相,流速：1.0 mL·min^-1,柱温：30℃,漂移管温度为105℃,N2流速（SPLM）2.85 mL·min^-1。结果：黄芪甲苷在0.007－0.140 mg/mL浓度范围内有良好线性关系（r=0.9996）,平均加样回收率为97.0%,RSD%为1.24%。结论：本方法灵敏度高、准确、重复性好、专属性强,可作为阿胶三宝膏质量标准提升的重要补充。Objective： To establish a HPLC-ELSD method for the determination of Astragaloside IV in Ejiao Sanbao decoction （阿胶三宝膏）. Methods： The HPLC system consisted of the FORTIS-C18 （4.6 mm×250 mm,5μm）column, the mobile phase consisted of acetonitrile （21）-0.05% KH2PO4 （79）, the flow rate was 1.0mL·min-1, the column temperature was 30℃, The temperature of drift tube was 105 ℃, SPLM was 2.85 mL·min^-1. Results： The linear response range was 0.007--0.140 mg/mL （r=0.9996）. The average recovery of astragaloside IV was 97.0%, and RSD was 1.24%. Conclusions： The method high sensitivity, accurate, repeatable and high specificity. It can be an important supplement of Ejiao Sanbao decoction elevated standard.
