兰地洛尔和艾司洛尔抗实验性心律失常作用和安全性评价的比较研究

摘　　要：目的比较兰地洛尔和艾司洛尔的抗心律失常作用和安全性。方法用序贯法测定兰地洛尔和艾司洛尔的半数致死量(LD50)、半数有效量(ED50),计算其治疗指数。用多种心律失常模型,观察比较兰地洛尔和艾司洛尔在等效剂量下的抗心律失常作用。结果兰地洛尔和艾司洛尔的ED50、LD50值分别为14.4,347.0 mg·kg-1和17.3,100.0 mg·kg-1,两药的治疗指数分别为24.1和5.8。兰地洛尔和艾司洛尔抗氯仿诱导的小鼠心律失常作用的等效剂量比为0.83∶1。两药对多种心律失常模型均有相似的抗心律失常作用,但艾司洛尔减慢心率的作用较强,而兰地洛尔的作用相对平缓。结论与艾司洛尔相比,等效剂量下的兰地洛尔具有相似的抗心律失常作用,起效更快,减慢心率作用更平缓,且治疗指数更高,更加安全。Objective To compare the antiarrhythmic effects and safety of landiolol and esmolol.Methods The sequential method was used to detect the median lethal dose（LD50） and 50% effective dose（ED50） of the two drugs.Treatment index were calculated by LD50/ ED50.Several of arrhythmia models were used to study the antiarrhythmic effects of landiolol and esmolol in the equivalent dose.Results ED50 and LD50of landiolol and esmolol were 14.4,17.3 mg·kg^-1and 347,100 mg·kg^-1respectively,TI were 24.1 and 5.8.The ratio of equivalent dose between landiolol and esmolol on anti-chloroform induced arrhythmia was0.83∶ 1.Landiolol and esmolol have similar anti-arrythmia effect on various arrhythmia models.Esmolol has a stronger inhibition function on the heart rate than that of landiolol.Conclusion Compared with esmolol,landiolol has similar antiarrhythmic effect,gentle and slow effect on heart rate,higher treatment index and safety in equivalent dose.
