摘 要:考察了不同提取方法对北五味子[Schisandra chinensis(Turcz.)Baill]多糖提取率的影响。分别采用回流法、微波辅助法、闪式法对北五味子多糖进行提取,通过可见-分光光度法测定其含量,设计响应面试验对其最佳方法的工艺进行优化。闪式法多糖提取率高于回流法和微波辅助法,其最佳工艺为料液比1∶20(m∶V),提取时间127 s、提取电压190 V、提取温度78℃,在此条件下北五味子多糖的提取率达到20.97%。不同提取方法对多糖的提取率影响较大,闪式法多糖提取率高,耗费时间短,溶剂用量少。The effects of different extraction methods on the yields of polysaccharide extracted from Schisandra chinensis(Turcz.) Baill were investigated. Reflux method, microwave-assisted method and smashing tissue method were used to extract Schisandra chinensis(Turcz.) Baill polysaccharides. Visible spectrophotometry was used to determine the yields of polysaccharide. Response surface methodology was used to optimize conditions of extraction. The yields of extraction obtained from smashing tissue technique was higher than that of reflux method and microwave-assisted method. The optimal conditions of extraction were the 1∶20 of solid-liquid ratio(m∶V), 127 s of extraction time, 190 V of extraction voltage and 78 ℃ of extraction temperature. Under these conditions, the yields of polysaccharide reached 20.97%. Extraction methods had great effects on yields of polysaccharide. Smashing tissue technique had the highest extraction yields with less time, energy and solvent.
