摘 要:目的:观察肝郁脾虚型大鼠海马及杏仁核中BDNF mRNA及蛋白表达变化,以及GluR1/2水平的变化,探讨其在肝郁脾虚型大鼠模型发病中的作用机制。方法:将45只Wistar大鼠随机分为3组:空白组、模型组和逍遥散组,每组15只,采用夹尾激怒+饮食失节法建立肝郁脾虚型大鼠模型,免疫组化S-P法和RT-PCR法检测各组大鼠海马及杏仁核中BDNF基因和蛋白表达,并采用ELISA法检测GluR1/2水平的变化。结果:模型组大鼠海马及杏仁核中BDNF mRNA和蛋白的表达与空白组比较均降低,经过逍遥散治疗后的BDNF表达量则有所升高(P<0.01,P<0.05);模型组GluR1升高GluR2降低,治疗后GluR1降低GluR2升高(P<0.05),差异均有显著性意义。结论:肝郁脾虚证的发生与BDNF的表达受抑密切相关,上调BDNF的表达有望成为肝郁脾虚证的治疗新方向。Objective:To study the dynamic expression of BDNF and Glu1 /2 in rat' s hippocampus and the amygdala of liver stagnation and spleen deficiency type and identify its underlying mechanism.Methods:45 rats were randomly divided into 3groups:the blank group,the model group and the Xiaoyao Powder group.Each group had 15 rats.The models were prepared by means of binding method and intemperance of taking food.We detected BDNF gene and protein expressions in rat's hippocampus and the amygdala by RT-PCR and Immunohistochemical method and detected GluR1 /2 by ELISA.Results:Compared with the blank group,the BDNF gene and protein expressions were decreased significantly in the model group and increased in the Xiaoyao Powder group(P〈0.01,P〈0.05).The GluR1 was increased and GluR2 was decreased in the model group.The GluR1 was decreased and GluR2 was increased in the Xiaoyao Powder group(P〈0.05).Conclusion:The occurrence of the liver stagnation and spleen deficiency type is closely related to the BDNF's inhibition of expression and up-regulating the expression of BDNF is expected to become a new way for the treatment of liver stagnation and spleen deficiency type.
