辛伐他汀对慢性阻塞性肺病模型大鼠的治疗作用及其机制研究

摘　　要：目的研究慢性阻塞性肺病(COPD)大鼠应用辛伐他汀治疗的作用机制。方法雄性健康SD大鼠60只随机分为研究组、模型组、对照组,每组20只,研究组、模型组大鼠建立COPD模型。2周后研究组予辛伐他汀灌胃,模型组、对照组予生理盐水灌胃,连续8周。观察3组肺泡灌洗液(BALF)细胞总数、分类计数水平、白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)浓度,血清IL-8、TNF-α、C反应蛋白(CRP)浓度。结果造模后COPD大鼠支气管管壁及血管周围均有多种炎性细胞浸润,肺泡壁变薄,大量融合形成大疱。3组BALF细胞总数、分类计数水平、IL-8、TNF-α浓度和血清IL-8、TNF-α、CRP浓度均有明显差异(P<0.05)。结论 COPD大鼠应用辛伐他汀治疗,可使BALF、血清中的炎症因子降低,减轻气道、肺组织炎症,从而达到治疗目的。Objective To study the mechanisms of Simvastatin in treatment of rats with COPD（ chronic obstructive pulmonary disease）. Methods A total of 60 healthy male Wistar rats were randomly divided into study group（ n =20）,model group（ n = 20） and control group（ n = 20）,and the COPD models were established in the study and model groups. After 2 weeks,the study group was lavaged with Simvastatin,while the model and control groups were lavaged with saline continuously for 8 weeks. The total cellular score,differential count level,IL-8（ interleukin-8）,TNF-α（ tumor necrosis factor-alpha） concentration of BALF（ bronchoalveolar lavage fluid）,and concentrations of serum IL-8,TNF-α and CRP（ C reactive protein） in the three groups were observed. Results After model establishment,a variety of inflammatory cell infiltrations were found in the bronchial wall and blood vessels,and the alveolar walls were made thin and fused into bulla. The differences in total cellular score,differential count level,IL-8,TNF-α concentration of BALF,and concentrations of serum IL-8,TNF-α and CRP in the three groups were statistically significant（ P〈0. 05）. Conclusion The Simvastatin in treatment of COPD rats can reduce inflammatory factors in BALF and serum and alleviate inflammation of the airways and pulmonary tissues,so that it may achieve the treatment result.
