摘  要:【目的】探讨缺硼导致樱桃萝卜细胞壁松弛的机理,为进一步丰富细胞壁的研究内容提供参考。【方法】以樱桃萝卜为试验材料,利用硼含量46.3μmol/L的营养液培养方法,以不含硼的蒸馏水作对照,通过HPLC法测定樱桃萝卜肉质根中的细胞壁结合硼含量,TBA法测定KDO含量,并用免疫化学方法定位细胞壁中的RG-II。【结果】缺硼处理导致樱桃萝卜肉质根中细胞壁结合硼的含量由14.21 mg/kg极显著下降到7.24 mg/kg(P<0.01),下降49.0%。细胞壁果胶多糖中的KDO含量变化不显著(P>0.05),缺硼处理与对照相比,免疫RG-II的荧光强弱和胶体金颗粒的密度无明显差别。【结论】樱桃萝卜缺硼抑制了其B-RG-II的形成,使细胞壁的稳定性降低而松弛。[Objective] The mechanism of cell wall loosening of cherry radish( Raphanus sativus L. var. radculus pers) because of boron deficiency was explored to provide references for enriching the research on cell wall. 【Method】Cherry radishes were grown in nutrient solution containing 46.3 μmol/L boric acid. The distilled water without boron was used as the control. Content of cell wall B of the fleshy roots was determined by HPLC method. KDO content was determined by TBA method,and RG-II of cell wall was located by immunochemical method. 【 Result 】 Due to the lack of B, the content of cell wall B decreased significantly by 49.0%, which was decreased from 14.21 to 7.24 mg/kg( P0.01),but the content of KDO were not influenced significantly. Fluorescence strength and granular density of RG-II showed no obvious difference between two treatments. 【Conclusion 】The formation of B-RG-II is inhibited because of B deficiency,and the stability of cell wall is reduced for the lack of B-RG-II,which leads to cell wall loosening.
