摘  要:目的探讨小菜蛾抗菌肽对人肝癌细胞株Hep G2增殖的抑制活性,合理制定小菜蛾的用量范围。方法建立小菜蛾抗菌肽系列样品溶液的HPLC指纹图谱,MTT比色法观察样品溶液对Hep G2细胞增殖的影响,绘制小菜蛾抗菌肽系列样品浓度C(mg·ml-1)与抑制率(%)之间的"量-效"曲线。结果 HPLC指纹图谱结果显示,不同浓度梯度的小菜蛾抗菌肽样品溶液的相似度良好;确定小菜蛾抗菌肽的最小有效用量30μg·ml-1,最大有效用量400μg·ml-1。结论小菜蛾抗菌肽对Hep G2细胞增殖抑制作用的合理用量范围为35.93μg^479.04μg。Objective To formulate quality standards of Plutella xylostella and observe the effect of pxCECA1 from Plutella xylostella( CA) on proliferation of HepG2 cells. Methods Different amounts of CA were added to the negative sample solution,anti-proliferative effects of a series of CA sample solution against HepG2 cells were evaluated by MTT assay in vitro,and a comparison between different series of CA was made. The HPLC fingerprint of a series of CA sample solution was calculated using the Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine( Version 2004A). Results There was much similarity between the series of CA samples according to the HPLC fingerprint. The minimum effective concentration of CA in Plutella xylostella was 30 μg·ml^-1while the maximum effective concentration was 400 μg·ml^-1. Conclusion The effective administration dosage of Plutella xylostella for restraining HepG2 cells proliferation ranges from 35. 92 μg to 479. 04 μg.
