摘 要:目的用银杏叶提取物温敏型鼻用原位凝胶治疗大鼠痴呆症,研究相关酶的活性和丙二醛(MDA)含量的改变。方法采用大鼠双侧颈总动脉结扎,建立大鼠痴呆模型。术后大鼠随机分为8组,即假手术组、模型组、空白对照组、阳性对照组(银杏叶分散片组)、银杏叶提取物温敏型鼻用原位凝胶低、中、高剂量组(6.25、12.5、25 mg·kg-1)和银杏叶提取物温敏型鼻用原位凝胶预防组,每组8只。术后鼻腔给药15 d,每天给药1次,末次给药3 h后,取血液、脑脊液和脑组织,检测超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、乙酰胆碱酯酶(ACh E)活性和MDA含量。结果模型组大鼠SOD、CAT活性明显降低,ACh E活性明显增强,MDA含量明显增加,与假手术组大鼠比较,有统计学差异(P<0.01,P<0.05);银杏叶提取物温敏型鼻用原位凝胶中、高剂量组SOD、CAT活性明显提高,ACh E活性明显减弱,MDA含量明显减少,与模型组比较,有统计学差异(P<0.01,P<0.05)。结论银杏叶提取物温敏型鼻用原位凝胶能明显提高SOD、CAT活性,降低ACh E活性,减少MDA含量,结果表明本制剂可能对痴呆症有一定治疗作用。Objective To study the related enzymatic activity and content changes of malondialdehyde( MDA) when gingkgo biloba extract temperature sensitive nasal in situ gel was used treatment of dementia in rats.Methods The rats were subjected to bilateral common carotid artery ligation to establish a senile dementia model.Sixty-four Wistar rats were randomly divided into eight groups: sham group,model group,blank control group,positive control group( gingkgo biloba dispersible tablet group),gingkgo biloba extract temperature sensitive nasal in situ gel 6. 25,12. 5,25 mg·kg^-1group and gingkgo biloba extract temperature sensitive nasal in situ gel prevention group. Postoperative nasal administration lasted 15 d,once a day. 3 h after the last administration,the SOD,CAT,ACh E activity and MDA content were examined in the plasma,cerebrospinal fluid and brain tissue. Results Compared with the sham group,the model group could significantly decrease SOD and CAT activity,significantly increase ACh E activity and MDA content( P〈0. 01,P〈0. 05). Compared with the model group,the 12. 5,25mg·kg^-1group could significantly increase SOD and CAT activity while significantly decreasing ACh E activity MDA content( P〈0. 01,P〈0. 05). Conclusion Gingkgo biloba extract temperature sensitive nasal in situ gel could significantly improve the activity of SOD and CAT,reduce ACh E activity and MDA content. It has been proved that the preparation is effective for dementia.
