摘 要:目的探讨骨髓间充质干细胞(BMCs)在去细胞神经修复大鼠坐骨神经损伤过程中对外周血调节性T淋巴细胞的影响。方法选择体质量为150~200 g、2月龄健康雄性SD大鼠15只及雄性Wistar大鼠15只。手术切取SD大鼠的坐骨神经以造成坐骨神经缺损,同时利用切除的神经分离雪旺细胞。手术获取15条Wistar大鼠的坐骨神经,化学萃取法制备去细胞神经,并根据需要向去细胞神经中植入雪旺细胞和BMSCs。SD大鼠随机分为3组:去细胞神经组,采用去细胞神经移植修复缺损;去细胞神经+雪旺细胞组,采用植入雪旺细胞的去细胞神经修复缺损;去细胞神经+雪旺细胞+BMSCs组,采用同时植入雪旺细胞和BMSCs的去细胞神经修复缺损。分别在移植前及移植后3、7、14 d用流式细胞仪检测各组SD大鼠尾静脉血标本中的T细胞亚群。多组间均数比较采用单因素方差分析,多组间两两比较及同组内多个时间点两两比较采用LSD-t检验。结果移植前去细胞神经组、去细胞神经+雪旺细胞组及去细胞神经+雪旺细胞+BMSCs组受体SD大鼠CD4+、CD25+及CD4+CD25+Foxp3+T细胞比例差异均无统计学意义(P均〉0.05)。移植后,各组3种T细胞比例均呈逐渐上升趋势,移植后14 d达到峰值,与移植前、移植后3 d及移植后7 d相比,差异均具有统计学意义(P均〈0.05)。移植后14 d,去细胞神经组CD4+、CD25+及CD4+CD25+Foxp3+T细胞比例分别为(39.9±1.9)%、(9.15±0.17)%及(2.37±0.09)%;去细胞神经+雪旺细胞组CD4+、CD25+及CD4+CD25+Foxp3+T细胞比例分别为(41.0±1.7)%、(10.11±0.15)%及(2.47±0.05)%;去细胞神经+雪旺细胞组+BMSCs组CD4+、CD25+及CD4+CD25+Foxp3+T细胞比例分别为(38.1±1.6)%、(8.19±0.13)%及(2.79±0.12)%,可见移植后14 d去细胞神经+雪旺细胞+BMSCs组的CD4+及CD25+T细胞比例均低于去细胞神经组和去细�Objective To investigate the changes of peripheral blood regulatory T cells caused by bone marrow derived mesenehymal stem cells (BMSCs) in sciatic nerve injury rats after nerve transplantation. Methods Fifteen SD and Wistar rats (weighted 150-200 g, aged 2 months) were selected. Sciatic nerve defect of SD rats were caused by surgical resection and the Schwann cells were obtained from these resected nerves. Fifteen sciatic nerves of Wistar rats were also resected and prepared into acellular nerves by chemical extraction. BMSCs or Schwann cells were injected into acellular nerves according to scheduled protocol. SD rats were randomly divided into 3 groups, including acellular nerves group, acellular nerves + Sehwann ceils group and acellular nerves + Schwann cells + BMSCs group. Before operation and 3, 7, 14 days after operation, the percentage changes of regulatory T cells in rats'peripheral blood were detected by flow cytometry. The statistical analysis was performed using one-way ANOVA and LSD-t. Results Before transplantation, the CIM+ , CD25+ and CD4 + CD25+ Foxp3 + T cells ratio showed no difference among acellular nerves group, acellular nerves + Schwann cells group and acellular nerves + Schwann cells + BMSCs group. Post- transplantation, the CD4 + , CD25 + and CD4 + CD25 + Foxp3+ T cells ratio presented an increasing tendency in SD rats and peaked on fourteen days after operation, compared with other time points, the differences were statistically significant ( P 〈 0. 05 ). Fourteen days after operation, in acellular nerve group, the CD4+ , CD25+ and CD4+ CD25+ Foxp3+ T cells ratio were (39.9 ±1.9)% , (9.15 ±0.17)% and (2.37 ±0. 09 )%, respectively. In acellular nerves + Schwann cells group, the CD4+, CD25+ and CIM + CD25 ~ Foxp3 + T cells ratio were (41.0 ± 1.7) %, ( 10.11 ± 0. 15 ) % and (2.47 ± 0. 05 ) %, respectively.
