栀子-连翘药对配伍的指纹图谱及主要化学成分研究

摘　　要：目的:建立栀子、连翘、栀子-连翘药对HPLC指纹图谱,研究栀子-连翘药对配伍前后指纹图谱变化情况,研究栀子-连翘药对配伍前后主要化学成分栀子苷、连翘苷的变化情况。方法:水煎煮制备栀子、连翘、栀子-连翘药对提取物,采用高效液相色谱法测定栀子、连翘单味药提取液及栀子-连翘药对提取液得到指纹图谱信息,使用中药指纹图谱软件处理(2004A版),测定栀子、连翘单味药提取液,栀子-连翘药对提取液中栀子苷和连翘苷的含量。结果:配伍后药对指纹图谱与栀子、连翘单药指纹图谱比较,药对指纹图谱的主要贡献来自连翘,二者有极好的相似度;药对指纹图谱与单药指纹图谱比较,基本为栀子、连翘两单味药特征峰的加和,没有明显新色谱峰的增加,配伍后仅特征峰2面积显著增加(P<0.05)。栀子苷、连翘苷提取量在单药煎液与药对煎液中变化不明显。结论:栀子连翘药对配伍前后指纹图谱未发生显著变化,栀子-连翘药对配伍前后主要指标性成分栀子苷、连翘苷未发生明显变化。Objective： To assess difference between the fingerprints of Fructus Forsythiae, Fructus Gardeniae, and the herb couple. Methods： The fingerprints of Fructus Forsythiae, Fructus Gardeniae, and the herb couple were detected by HPLC, and the data were processed with the fingerprint software （2004, Edition A）. The contents of geniposide and phillyrin were determined. Results： The fingerprints of the herb couple were closer to that of Fructus Forsythiae than to the fingerprint of Fructus Gardeniae. The peaks of the herb couple were consistent with those of the two herbs and no new fingerprint was added except that the area of peak 2 of the herb couple ex- panded significantly （P 〈 0.05）. The ratio of geniposide in the extracts of Fructus Gardeniae and the herb couple was 24.43 to 23.86, and the ratio of phillyrin in Fructus Forsythiae and the herb couple is 2.64 to 2.63. Conclusion： No significant difference has been found between the fingerprints of the two single herbs and the herb couple. No significant difference exists between the contents of geniposide and phillyrin in the extracts of Fructus Forsythiae, Fructus Gardeniae and the herb couple.
