摘  要:目的观察丹参水提物对大鼠离体心脏缺血再灌注损伤的保护作用。方法采用大鼠离体心脏灌流技术制备心脏缺血再灌注损伤模型,用不同浓度丹参水提物灌流心脏。观察记录复灌50 min时的心肌功能变化。结果丹参水提物可提高心脏缺血再灌期的左心室舒张压(LVDP)、左心室内压最大上升速率(+dp/dtmax)、左心室内压最大下降速率(-dp/dtmax)水平,差异具有统计学意义(P<0.01),降低左心室舒张末压(LVEDP)、肌酸激酶(CK)和乳酸脱氢酶(LDH)水平,差异具有统计学意义(P<0.01),提高心肌中p-Akt水平,差异具有统计学意义(P<0.01)。结论丹参水提物对缺血再灌注损伤心脏具有保护作用,这一保护作用与它调节磷脂酰肌醇-3-激酶/丝苏氨酸蛋白激酶(PI3K/Akt)信号通路有关。Objective To observe the protective effect of Salvia Miltiorrhiza aqueous extract for mice isolated heart ischemia reperfusion.Methods Perfusion technique of isolated heart was applied to make isolated heartischemia reperfusion model, and different concentrations of Salvia Miltiorrhiza aqueous extracts were used for reperfusion. Records were made on the changes of myocardial function at 50 min of reperfusion. Results Salvia Miltiorrhiza aqueous extract could improve levels of LVDP, left heart interior maximum pressure rise rate (+dp/dtmax), left heart indoor pressure maximum decline rate (-dp/dtmax) during heart ischemia reperfusion, and the difference had statistical significance (P〈0.01). Levels of LVEDP, CK, and LDH could be reduced, and the difference had statistical significance (P〈0.01). Level of p-Akt in myocardium could also be improved, and the difference had statistical significance (P〈0.01).Conclusion Salvia Miltiorrhiza aqueous extract provides protective effect for isolated heart ischemia reperfusion, and its effect is related with its regulation of PI3K/Aktsignal channel.
