摘  要:目的:观察一次性力竭运动过程中大鼠苍白球(GP)胞外GABA、Glu浓度及GABAARα1与GluR2表达的变化规律,揭示GP在运动性疲劳中枢调控中的作用机制。方法:Wistar雄性大鼠随机分为神经递质浓度测定组和受体表达测定组,受体测定组又分安静、疲劳和恢复3个亚组。采用微透析-高效液相色谱联用技术,实时在线检测大鼠一次性力竭运动过程中GP胞外GABA、Glu浓度的动态变化规律;采用免疫荧光技术观察不同运动阶段大鼠GP内GABAARα1与GluR2的表达情况。结果:一次性力竭运动过程中大鼠GP胞外GABA、Glu浓度呈波浪式变化规律。大鼠运动至力竭时GP胞外GABA、Glu浓度及GABAARα1与GluR2表达较安静时显著升高(P<0.01)。Glu/GABA比值在运动过程中出现两次波峰,且与大鼠的运动行为表现相一致。结论:GABA、Glu分别通过GABAARα1与GluR2共同参与了一次性力竭运动过程中大鼠GP神经元兴奋性的调节。由此推测大鼠GP胞外GABA大量堆积和GABAAR表达上调,过度激活了间接通路,可能是导致运动疲劳产生的主要原因之一。Objective: To investigate the concentration of GABA and Glu and the expression of GABAARα1and GluR2 in rat globus pallidus( GP) during one- time exhaustive exercise and reveal the mechanism of GP in the neural regulation of exercise- induced fatigue. M ethods: Wistar rats w ere randomly divided into tw o groups of neurotransmitter concentration and receptor expression. The second group w as divided into three subgroups of resting,fatigue and restoring. M icrodialysis-HPLC w as applied for real- time online detection of concentrations of GABA and Glu. Immunofluorescence technique w as applied for observation of expression of GABAARα1and GluR2 in three stages( resting,fatigue and restoring state). Results: The concentration of GABA and Glu show ed w avy curves during one- time exhaustive exercise. Compared to that of resting state,the concentration of GABA and Glu and the expression of GABAARα1and GluR2 at fatigue state w ere significantly increased( P 0. 01). The ratio of Glu / GABA had tw o peaks in the process of movement,and w as accorded w ith the motor behavior of rats. Conclusion: GABA and Glu w ere involved in the regulation of excitability in rat GP neurons by GABAARα1and GluR2 respectively during one- time exhaustive exercise. Accumulation of extracellular GABA and up-regulation of GABAAR in rat GP activate indirect pathw ay,w hich may be one of central mechanisms of exercise- induced fatigue.
