摘 要:采用硫酸-苯酚法测定红蓝草多糖的含量,用正交设计法筛选和优化红蓝草多糖的提取工艺。以料液比、提取时间、溶液的p H值、提取温度作为因素,进行L9(34)正交试验。各因素对提取红蓝草水溶性多糖的影响为:提取温度>料液比>提取溶剂p H值>提取时间。确定较优提取工艺为料液比为1:40(m:v),提取温度100℃,提取时间为2h,提取溶剂p H值4.5,在该工艺条件下,红蓝草多糖的得率为8.018%,说明红蓝草除了主成分紫蓝素类、黄酮类外,其多糖的含量也较高。To optimize the extraction process of polysaccharide in Peristrophe roxburghiana. Sulfuric acid- phenol method is used to determine the content of polysaccharide in Peristrophe roxburghiana. The influence of solid- liquid ratio、temperature、 time and extraction solution p H on extraction yield of the polysaccharide in Peristrophe roxburghiana were discussed with orthogonal test method. The impact sequence of the factors on the extraction rate of polysaccharide in Peristrophe roxburghiana was as follows: temperaturesolid-liquid ratioextraction solution p Htime; The optimal extraction condition was extraction temperature of 100℃、solid- liquid ratio of 1:40、extraction time of 2h and extracted solution p H=4.5. Under these optimal conditions,the extracting rates of polysaccharide in Peristrophe roxburghiana is 8.018%.The content of polysaccharide is high which can be exploited and utilized as another new ingredient.
