摘 要:目的探讨维吾尔族(简称维族)与汉族Graves病(GD)患者血清白细胞介素17(IL-17)与IL-17rs763780基因多态性的差异及其与CD之间的关系。方法选择初发、未经治疗的GD患者193例(GD组,维族99例,汉族94例)及健康体检者188例(健康对照组,维族97例,汉族91例),应用酶联免疫吸附法检测血清IL-17水平。用聚合酶链反应-限制性片段长度多态性分析方法 (PCR-RFLP)测定IL-17rs763780基因多态性。结果与健康对照组比较,GD组血清IL-17水平明显升高,差异有统计学意义;维族GD患者血清IL-17水平较汉族GD患者明显升高,差异有统计学意义。GD组血清IL-17水平与TRAb、FT3、FT4呈正相关(r=0.139,P<0.05;r=0.159,P<0.05;r=0.047,P<0.01)。与健康对照组比较,GD组IL-17rs763780等位基因T分布高于等位基因C,差异有统计学意义。与健康对照组比较,GD患者IL-17rs763780位点TT分布高于TC、CC基因型,差异有统计学意义(P<0.05)。与汉族GD组比较,维族GD患者IL-17rs763780等位基因C和T分布比较差异无统计学意义;维族GD患者与汉族GD患者IL-17rs763780位点CC、TC、TT基因型分布比较差异无统计学意义(P>0.05)。GD组3种不同基因型(CC、TC、TT)血清IL-17水平有差异。结论 IL-17在GD中的高表达提示其与可能与GD的发病相关;IL-17rs763780基因多态性可能与IL-17水平有关。Objective To explore the differences of serum interleukin-17 (IL-17)and IL-17 rs763780 gene polymorphism between Uyghur and Han GD patients,and the relationship with Graves disease (GD). Methods Choose the incipient,193 patients was untreated GD (GD group,99 of Uyghur and Han,94 ca-ses)and 188 cases of healthy physical examination(healthy control group,97 cases Uyghur and han,91 cases),using enzyme-linked immunosorbent method to detect the serum level of IL-17.Using polymerase chain reaction-restriction fragment length polymorphism analysis method (PCR-RFLP)determination of IL-17 rs763780 gene polymorphism.Results GD group compared with control group,the serum IL-17 level increased significantly,the difference was statistically significant,Uyghur compared with Han pa-tients with GD,GD patients serum levels of IL-17 Uyghur patients increased significantly ,the difference was statistically significant.In the hyperthyroidism group,the serum level of IL-17 and TRAb,FT3,FT4 were positively correlated (r =0.139,P 〈 0.05;r =0.159,P 〈 0.05;r =0.047,P 〈0.01).Compared with healthy controls,patients with GD group of IL-17 rs763780 allele T distribution is higher than the distri-bution of allele C,the difference was statistically significant.Compared with healthy controls,patients with GD IL-17 rs763780 loci TT genotype distribution above TC,CC genotype,the difference was statisti-cally significant (P 〈0.05).To compared with the Han GD group,there was no statistically significant difference in Uyghur GD group of IL-17 rs763780 allele C and T distribution;Uyghur GD patients com-pared with Han GD patients ,there was no statistically significant difference in genotype distribution of IL-17 rs763780 loci CC,TC,TT (P 〉0.05).GD group 3 different genotypes (CC,TC,TT)between the serum level of IL-17.Conclusion High expression of IL-17 in GD prompt its and may be associated with the onset of GD;IL-17 rs763780 gene polymorphism may be related to the level of IL-17.
