摘 要:为明确山东地区葡萄霜霉病菌株的致病性及亲缘关系,本研究以不同抗性的6个葡萄品种作为鉴别寄主,采用叶盘接种法,对采自山东地区的13个葡萄霜霉病菌株进行致病性测定,并扩增每个菌株的r DNA-ITS序列进行同源性分析。结果表明,这13个葡萄霜霉病菌株均有致病性,据致病力分为强、中、弱3种致病类型,中等致病型菌株占优势,菌株间致病力差异与菌株的地域来源无明显相关性。序列比对及系统发育树分析表明,这13个葡萄霜霉病菌株的亲缘关系很近,r DNA-ITS序列同源性高达99.94%,菌株间致病力差异与r DNA-ITS序列多态性不相关。In order to study the virulence differentiation and genetic relationship of Plasmopara viticola in Shandong,13 different strains of P.viticola were collected from different regions in Shandong Province.Their pathogenicity were tested through infecting 6 grape varieties with different resistance by inoculating leaf disc method,and their homology were analyzed by amplifying the rDNA -ITS sequences with primers ITS1F/ITS4R.The results indicated that all the 13 strains had pathogenicity.The cluster analysis concluded that they were roughly divided into weak, moderate and strong pathotypes, and the moderate pathotype was dominant. The pathogenicity differentiation was not obviously correlated with the regional origin.The sequence alignment and phylogenetic tree analysis results showed that there were very close genetic relationships between the 13 strains with the homolygy of rDNA-ITS sequences as 99.94%;moreover,there was no correlation between virulence differentiation and rDAN-ITS sequences polymorphism.
