摘  要:目的利用中药治疗机理,根据蛋白质组学的研究方法对激素性骨坏死的病理进行研究,为临床治疗激素性骨坏死提供一些参考。方法建立常用激素性骨坏死和中医治疗激素性骨坏死的大鼠模型对照组,并设立一组空白对照组,4周后根据骨形态学方法鉴定模型,确认成功后提取大鼠骨组织蛋白质样品,运用蛋白质组学分析方法,针对得到的数据和图谱进行进一步的研究。结果三组大鼠骨组织总蛋白质分子的解剖图谱和蛋白质肽指纹图谱通过蛋白质组学分析发现,有三种蛋白质存在明显差异。结论在激素性骨坏死发病中发挥作用的阻凝蛋白重链ⅡB、磷脂谷胱甘肽过氧分酶及泛素化酶E2,亦影响了中药对激素性骨坏死的治疗结果,有着重要的调控作用。Objective Using the mechanism of traditional Chinese medicine treatment, according to the research of the proteomic approach, to study of hormone osteonecrosis of pathology, to provide some references for the clinical treatment of glucocorticoid-induced osteonecrosis. Methods Established two rats models groups, one control group with common hormone osteonecrosis and TCM treatment of glucucorticoid-induced osteonecrosis, another control group with blank. After four weeks, identified model based on bone morphology method after the success of the confirmation from rat bone tissue protein samples, used proteomics analysis method according to the data and map for further research. Results Analyzed three groups of rats bone tissue total protein molecular anatomy atlas peptide and protein fingerprint by proteomic. There were obvious difference among three kinds of protein. Conclusion Anticoagulating Ⅱ B heavy chain of protein, lecithin glutathiune oxygen enzyme and enzyme E2 ubiquitin play important regulatory role in the incidence of glucocorticoid-induced osteonecrosis and influence the treatment results of traditional Chinese medicine treatment of glucocorticoid- induced osteonecrosis.
