摘 要:目的:观察经阴道注入重组白细胞介素17(r IL-17)对正常妊娠CBA/J孕鼠(CBA/J雌鼠×BALB/c雄鼠)妊娠结局的影响,探讨其在诱导母胎免疫排斥中的作用。方法:将6-8周龄清洁级CBA/J(雌)和BALB/c(雄)近交系小鼠分为3组。对照组:正常妊娠模型CBA/J孕鼠未作任何干扰。磷酸盐缓冲液(PBS)组:正常妊娠模型CBA/J孕鼠,于受孕第1天麻醉状态下经阴道注射50μL PBS。r IL-17组:正常妊娠模型CBA/J孕鼠,于受孕第1天麻醉状态下经阴道注射50μL r IL-17。受孕后第14天观察胚胎丢失率。通过实时荧光定量聚合酶链反应(q RT-PCR)方法、蛋白质印迹(Western blotting)方法检测鼠蜕膜转化生长因子β(TGF-β)和IL-10的表达。结果:给予正常妊娠模型孕鼠外源性r IL-17,能引起其胚胎丢失率明显增加,且蜕膜TGF-β和IL-10 m RNA及蛋白的表达均明显降低(P〈0.05)。而对照组与PBS组相比,胚胎丢失率及TGF-β和IL-10 m RNA及蛋白的表达差异均无统计学意义(均P〉0.05)。结论:经阴道注入r IL-17能够引起正常妊娠模型CBA/J孕鼠胚胎丢失率增加,并引起母胎界面免疫耐受细胞因子分泌异常。Objective: To investigate the effect of transvaginal injected recombinant IL-17 on the pregnancy outcome in the CBA/J mice model (CBA/JxBALB/c) and its role in feto-maternal immune rejection. Methods:Six to eight-week-old female CBA/J and male BALB/c were divided into three different experimental groups. Mice of normal pregnancy (CBA/Jx BALB/c ) received no treatment were served as control group. 50 μL phosphate-buffered saline (PBS) or 50 p.L recombinant IL-17 (rIL-17) was administered in the vaginal vaults of anesthetized pregnant CBA/J mice on Day 1 of pregnancy served as PBS group and rIL-17 group individually. The numbers of surviving and reabsorbed fetuses in each group were counted, and the expression of TGF-β and IL-IO in decidua was assessed by quantitative real-time PCR and western blotting on Day 14 of pregnancy. Results:In this in vivo model of normal pregnancy, exogenous rIL-17 resulted in increased spontaneous resorption of fetuses and decreased expression of TGF-β and IL-10 (P〈0.05). And there was no statistical difference between PBS group and control group (P〉0.05). Conclusions :Intravaginal rIL-17 can induce inflammation in fetomaternal interface resulting in abortion in normal pregnant CBA/J mice model.
