眼针运动疗法对脑缺血再灌注大鼠脑缺血半暗带区VEGF蛋白及VEGFmRNA表达的影响

摘　　要：目的:观察眼针运动疗法对脑缺血再灌注(MCAO)模型大鼠大脑缺血半暗带区域脑组织内VEGF、VEGFR1、VEGFR2表达的影响。方法:采用随机数字表法将SD大鼠按随机数字表法分为空白对照组、假手术组、模型复制组。对模型复制组大鼠采用线栓法进行脑缺血再灌注模型复制,再将模型复制组按随机数字表法分为模型对照组、眼针组、眼针加运动组,分别进行眼针、眼针运动疗法干预治疗,采用RT-PCR及western-blot法对各组大鼠脑组织中VEGF、VEGFR1/2基因及蛋白表达水平进行检测。结果:与空白对照组比较,模型对照组、眼针组及眼针运动疗法组大鼠脑缺血半暗带区域组织中VEGF、VEGFR1、VEGFR2 mRNA及蛋白表达水平显著升高;与模型对照组比较,眼针组及眼针运动疗法组大鼠脑缺血半暗带组织中VEGF、VEGFR1、VEGFR2mRNA及蛋白表达水平显著升高;眼针运动疗法组大鼠脑组织中VEGF、VEGFR1、VEGFR2 mRNA及蛋白表达水平高于眼针组。结论:眼针运动疗法能够促进脑缺血再灌注损伤后脑内缺血半暗带区域内VEGF、VEGFR1、VEGFR2 mRNA及蛋白的表达,从而促进半暗带组织中血管的新生,加快侧枝循环的建立,实现其脑保护作用。Objective： to observe the needle eye exercise therapy on cerebral ischemia reperfusion （MCAO） model in the rat brain ischemia half dark area within the brain tissue VEGF, VEGFR1, VEGFR2 expression. Methods： using random number method, the SD rats were randomly divided into blank control group, control group, the model copy group. Copy the model group rats by using line switch on cerebral ischemia reperfusion model replication, and then copy model group were randomly divided into model control group, the eye, eye needle plus exercise group. Respectively, eye for eye needle exercise therapy intervention treatment, by the method of Rt-pcr and western blot - of VEGF, groups of rats had VEGFR1/2 for testing the level of gene and protein expression. Results： compared with blank control group, model control group and eye, eye needle group exercise therapy in ischemia rats half dark with VEGF in regional organizations, VEGFR1, VEGFR2 mRNA and protein expression level increased significantly ; Compared with model control group, group and eye eye needle exercise therapy group of VEGF in ischemia rats half dark band organization, VEGFR1, VEGFR2 mRNA and protein expression level increased significantly; Needle eye exercise therapy in VEGF group rats had, VEGFR1, VEGFR2 mRNA and protein expression level is higher than eye needle group. Conclusion ： pay close attention to again needle eye exercise therapy can promote brain ischemia injury after brain ischemia within half dark belt area of VEGF, VEGFR1, VEGFR2 mRNA and protein expression, thus facilitating half dark with tissue, new blood vessels to speed up the establishment of collateral circulation, achieve the cerebral protective effect.
