三聚氰胺的FC数据拟合与ELISA试剂盒软件计算问题分析

摘　　要：通过对大量生鲜乳三聚氰胺试剂盒检测结果分析,明确了针对FC仪器需使用曲线跨度较小试剂盒。在曲线拟合中以试剂盒曲线、FC仪器中0点作为曲线值布点舍去非正常点后进行拟合较相似。0点作为曲线布点较试剂盒曲线CS拟合加标回收率、样品含量高;2种拟合方式三聚氰胺含量在0～5、5～ 10和10～ 20 μg/kg范围时样本数量差异不大。0点布成空白点曲线线性较好,但99％检测样品含量值显示“非数据”;除6、7、8曲线样品加标回收率偏低;0点赋微小值后进行曲线点板,除7、8曲线除外其余曲线R2符合试验要求,其加标回收率偏高。结论：试验中使用FC仪器,可选择曲线点位跨度较小试剂盒;若选用勤邦试剂盒、检测样品较多时则以0点布点舍去异常点后拟合计算。Based on the analysis of detection results of lots of fresh and raw milk by melamine kit,the kit with less span curve is suggested to use for FC instrument. During curve fitting,it is similar to fit curve by stationing zero in kit curve and FC instrument and rounding the abnormal point. The adding standard recovery with zero point as stationing is higher than that with kit curve CS. The numbers of samples within the range of 0 ～ 5 μg,5 ～ 10 μg,and 10 ～ 20 μg of melamine were less different between the two fitting methods. Linear of curve with zero stationing as blank point was better,but 99% samples tested indicated＂non-data＂except that the adding standard recovery was low in the 6,7 and 8 curve. When zero point bestow on little value,R2 was suitable for acquirement of experiment except for the 7 and 8 curve,but the adding standard recovery was higher. The results indicated that the kit with less span curve should be used for FC instrument. If Qinbang kits are selected and there are more detection samples,stationing zero rounding the abnormal point should be used to fit the curve.
