摘 要:目的研究高脂饮食喂养诱导的胰岛素抵抗(IR)小鼠骨骼肌中磷脂酰肌醇3激酶(P13K)表达及肌糖原含量的改变。方法将20只雄性KM小鼠随机分为对照组(NC组,10只)和IR组(10只),NC组予常规饲料,IR组予高脂饲料。16周时IR造模成功,记录小鼠体质量、空腹血糖(FBG)和空腹胰岛素(FINS),并计算稳态模型胰岛素抵抗指数(HOMA-IR);Westernblot法检测骨骼肌P13K的表达,过碘酸雪夫(PAS)染色观察骨骼肌糖原含量的变化。结果与NC组比较,IR组小鼠体质量、FBG、FINS及HOMA—IR增高(P均〈0.05),IR组骨骼肌中P13K的表达显著降低(P〈0.05);PAS染色显示NC组骨骼肌细胞内紫红色着色广泛,着色颗粒较多,IR组着色浅,着色颗粒较少。结论骨骼肌P13K表达降低可能是导致肌肉IR产生的原因之一。Objective To study the changes in phosphatidylinositol 3-kinase (PI3K) expression and glycogen content in the skeletal muscle of mice with insulin resistance(IR) fed with high fat diet. Methods 20 male KM mice were randomly divided into normal control group( NC group,n = 10) and IR group(n = 10). The mice in NC group were fed with routine diet and the mice in IR group were fed with high-fat diet. At the I6th week, IR models were tested successful. Record body weight, fasting blood glu- cose(FBG) and fasting insulin(FINS) ,then calculated Homeostasis Model Assessment for Insulin Resist- ance(HOMA-IR). The expression of PI3K in the skeletal muscle were measured by Western blotting method and the muscle glycogen content were measured by Periodic Acid-Schiff Stain (PAS). Results Compared with NC group, the body weight, FBG, FINS and HOMA-IR of IR group were higher(respective- ly P 〈 0.05 ), the PI3K in the skeletal muscle of IR group decreased significantly( P 〈 O. 05 ). The skeletal muscle cells in NC control were colored extensively and more colored granules,while IR gruop were col- ored light and less colored granules. Conclusion The reduced expression of PI3K in skeletal muscle may be one of the reasons leading to muscle IR.
