摘 要:目的探讨丹蛭降糖胶囊保护糖尿病大鼠肾脏的可能作用机制。方法 55只大鼠除正常组(9只)外,其余均给予高脂饲料喂养4周,按35 mg/kg给大鼠腹腔注射链脲佐菌素(STZ)1次建立糖尿病模型,造模成功的41只大鼠随机分为模型组11只,吡格列酮组和丹蛭高、低剂量组各10只。丹蛭高、低剂量组分别给予丹蛭降糖胶囊1.08、0.54 g/(kg·d)灌胃,吡格列酮组给予吡格列酮片10 mg/(kg·d)灌胃,模型组、正常组给予生理盐水5 ml/(kg·d)灌胃。连续灌胃8周,测定血糖,血脂,留取肾脏,做肾脏病理及肾组织骨桥蛋白(OPN)分析。结果模型组大鼠肾脏受损较重,而丹蛭高剂量组肾脏病变不显著,肾小球、肾小管结构基本清晰,丹蛭高剂量组病理改变程度轻于丹蛭低剂量组。模型组大鼠血糖、血脂水平及肾组织中OPN表达较正常组明显升高(P<0.01);丹蛭高、低剂量组及吡格列酮组大鼠血糖、血脂水平和肾组织OPN表达较模型组明显降低(P<0.05或P<0.01)。结论丹蛭降糖胶囊可明显改善糖尿病大鼠肾脏病变程度,有良好的降低血糖、调节血脂的疗效,可能与其下调肾组织OPN的高表达有关。Objective To discuss possible (DZJT capsule) for diabetic rats. Methods mechanism of kidney-protecting effect of Danzhi Jiangtang Capsule Of 55 rats, 9 were assigned into normal group, the other 46 rats were fed with high-fat diet for four weeks, then intraperitoneally injected streptozocin (STZ) 35mg/kg once to make model diabetes. Forty-one successful models were randomized into model group (n = 11 ), pioglitazone group (n = 10), low-dose DZJT capsule group (n = 10) and high-dose DZJT capsule group ( n = 10). The high-dose and the low-dose DZJT capsule group were respectively given DZJT capsule 1.08 g/ ( kg ·d), 0. 54 g/ ( kg ·d) as gavage. The pioglitazone group was given pioglitazone lOmg/ (kg ·d) by gavage, while the model group and the normal group were given normal saline 5 ml/ ( kg · d) by gavage, lasting for eight weeks. Then measured blood glucose and blood fat, and collected kidney tissue to analyze pathology and osteopontin (OPN). Results Kidney damaged seriously in the model group, but in the high-dose DZJT capsule group, no significant change was found, with the structure of kidney tubules and glomeruli legible. Pathologic c dose group. The level of blood glucose, blood fat hanges of the high-dose DZJT capsule group were less than the low-and OPN in the model group increased more obviously than that of the normal group (P 〈 0. 01 ). In the high-dose and the low-dose DZJT capsule group and pioglitazone group, the levels of blood glucose, blood fat and OPN decreased more obviously than in the model group ( P 〈 0. 05 or P 〈 0. 01 ). Conclusion DZJT capsule may markedly improve renal lesions of diabetic rats, effectively reduce blood glucose and blood fat. These effects may be due to DZJT capsule reducing the overexpression OPN in nephridial tissue.
