复方绞芪方对非酒精性脂肪性肝炎大鼠脂质代谢变化的影响

摘　　要：目的:观察复方绞芪方对高脂高糖饮食诱导的非酒精性脂肪性肝炎(NASH)大鼠脂质代谢变化情况的影响,探讨其防治NASH的作用机制。方法:高脂高糖饮食喂养12周诱导SD大鼠建立NASH模型,同时以不同剂量的复方绞芪方治疗,检测大鼠血清TG、CHOL、HDL-C、LDL-C、FFA、ALT、AST和肝组织匀浆TG、CHOL含量变化,常规HE染色观察肝组织的病理改变并计算NAFLD活动度积分(NAS)。结果:模型组大鼠的血清ALT、AST、LDL-C、FFA水平以及肝组织中的CHOL、TG含量较正常组明显增加;而血清HDL-C水平明显下降;NAS计分明显升高。应用复方绞芪方进行干预后,大鼠NAS计分较模型组大鼠显著降低,肝功能明显改善,血清CHOL、TG、LDL-C、FFA水平明显下降同时HDL-C明显升高,肝组织中CHOL和TG含量也明显下降。结论:高脂高糖饮食诱导的NASH大鼠存在脂质代谢紊乱,复方绞芪方能一定程度纠正肝组织脂质代谢紊乱,这可能是其防治NASH的重要机制。Objective：To investigate the method of Chinese Herbs with Fu Fang Jiao Qi Fang on studying the alteration of lipid in Rats during the progression of Non - Alcoholic Steatohepatitis,which was established by gavage of high fat and sugar diet. Method：The model of non - alcoholic Steatohepatitis was induced by high fat and sugar diet to SD rats for twelve weeks, and treated by different doses of Chinese medicine compound with functions of Fu Fang Jiao Qi Fang. De- termination of rat serum TG, CHOL, HDL - C, LDL - C, FFA , ALT, AST content and liver tissue TG, CHOL content change. The pathological changes of liver tissue were observed with HE staining to calculate NAFLD activity score. Result ： In model group, serum ALT ,AST, LDL - C ,FFA levels and liver tissue CHOL, TG levels were significantly increased, se- rum HDL - C level was decreased and the NAFLD activity scores of rats were significantly higher compared with normal. The NAFLD activity score, serum ALT, AST, TG,CHOL,LDL - C,FFA levels and liver tissue CHOL, TG levels in Chi- nese medicine compound groups were all improved lower that of model, HDL - C level was increased. Conclusion ： High fat and sugar diet - induced existence lipid metabolism in rats with Non - Alcoholic Steatohepatitis, Chinese Herbs with Fu Fang Jiao Qi Fang can correct to some extent liver lipid metabolism disorders, it may be the important mechanisms to Prevent and treat NASH.
