摘 要:目的探讨氢气对低压低氧环境中雄性Wistar大鼠生殖系统损伤的防护效果及作用机制。方法 48只大鼠随机分为常氧对照组(C组),低压低氧模型组(H组),低氧生理盐水干预组(HS组),低氧氢气饱和生理盐水干预组(HH组),各组又分为15 d组和30 d组,共计8组。用低压氧舱模拟海拔6 000 m环境,HS组腹腔注射生理盐水,HH组注射氢气饱和生理盐水。实验结束后,观察睾丸组织形态改变,检测精子密度等指标和性激素水平以及血清、睾丸组织活性氧和抗氧化酶活性。结果 H 15 d组和H 30 d组大鼠精子密度、活力、存活率均低于C组(P<0.05),而HH组大鼠精子密度和活力均高于H组和HS组(P<0.05)。HH 15 d和30 d组大鼠血清·OH低于同期H组(P<0.05),HH 30 d组睾丸ROS低于H 30 d组(P<0.01)。结论氢气对低压低氧环境造成的大鼠生殖损伤具有有效的防护效果。Objective To study the effect of hydrogen on reproductive function of male rats in hypoxia environment and its mechanism.Methods Forty-eight male Wistar rats were divided into 4 groups at random:blank control (C)-group,hypobaric hypoxia (H)-group,hypobaric hypoxia with physiological saline (HS)-group,and hypobaric hypoxia with saturated hydrogen and saline (HH)-group.All the rats were raised in low pressure oxygen cabin simulating 6 000 m environment except C-group.HS-group was injected with physiological saline[10 ml/(kg · d)],and HH-group was injected with saturated hydrogen saline [10 ml/(kg/d)].Samples were collected on the 15th day and the 30th day for the detection of sperm density,vitality,survival rate and abnormal rate and for the detection of serum testosterone (T),lutcinizing hormone (LH),follicle stimulating hormone (FSH) and other sex hormones,as well as reactive oxygen species (ROS) and total antioxidant enzyme capacity (T-AOC) in sera and testicular tissue.Results In hypobaric hypoxia conditions,sperm density,total energy and total survival rate were significantly lower in H-group than in C-group (P < 0.05).Sperm density and total energy were significantly higher in HH-group than in H-group or HS-group (P < 0.05).Hydroxyl radical in serum was significantly lower in HH-group than in H-group or HS-group (P < 0.05).Testicular reactive oxygen species (ROS) was significantly lower in the 30-day subgroup of HH-group than in that of H-group (P <0.01).Conclusion Hydrogen can effectively remove part of oxygen free radicals in rats,and protect reproductive function from suffering from hypobaric hypoxia induced injury.
