摘 要:目的:观察大鼠脊神经后根中枢突切断后相应背根神经节(DRG)卫星细胞(SGCs)Slit1的表达变化。方法:24只成年SD大鼠随机分为1、3、7d和14d组(n=6),脊神经L5、6左侧后根中枢突切断后,应用免疫组织化学方法检测各组相应DRG内卫星细胞Slit1表达变化。结果:DRG中枢突切断后1d组表达slit1阳性卫星细胞围绕的神经元比例开始增加,7d达到高峰,14d后逐渐下降,各组与对照组(右侧)比较有明显差异(P<0.01),各对照组间无明显差异(P>0.05)。对照组少量神经元slit1弱表达,1、3d组Slit1阳性的神经元比例分别为40.12%和59.45%,7、14d组神经元表达Slit1显著减少。结论:成年大鼠脊神经中枢突切断后,Slit1在SGCs和感觉神经元表达上调,SGCs源性Slit1可能参与了感觉神经损伤后修复过程。Objective:To observe changes of Slitl expression in satellite glial cells(SGCs) of rat dorsal root ganglion (DRG) after dorsal rhizotomy. Methods.. A total of 24 adult SD rats were randomly divided into 1 d,3 d,7 d and 14 d according to the survival time. After L5.6 dorsal rhizotomy, expression of Slit1 in the SGCs of DRG were detected respectively by immu- nohistochemical staining methods. Results= The rate of neurons surrounded by Slit-positive SGCs in ipsilateral DRG was in creased at 1 d after dorsal rhizotomy,reaching its peak at 7 d and decreased at 14 d. There was significant difference compared with control groups (right side,P〈0.01) ,but no difference among control groups (P〉0.05) . In 1 d,3 d groups ,the rate of Slit-positive-neurons in ipsilateral DRG was 40.12% and 59.45 %, respectively,the rate was significantly decreased in 7 d, 14 d groups, in contrast to slit1 weak expression in the control groups. Conclusions: The expression of Slitl in SGCs and neurons of DRG can he up-regulated by dorsal rhizotomy. Slitl from SGCs may participate in the process of regeneration after sensory nerve injury.
