摘 要:目的:研究绞股蓝总苷(Gypenosides,GP)对胆固醇所致人血管平滑肌细胞(Human Vascular Smooth Muscle Cells,HVSMCs)表型转化的影响。方法:体外培养人VSMCs株,分别用1.0、10.0、100.0μg·m L-1的GP预处理1 h后,再加入50.0 mg·L-1的胆固醇处理48 h。透射电镜观察细胞超微结构改变;Western blot法检测VSMCs中α-平滑肌肌动蛋白(Smooth Muscle alpha-Actin,α-SMA)、平滑肌22α(Smooth Muscle 22 alpha,SM22α)及表皮调节素(Epiregulin)蛋白表达;实时定量PCR检测VSMCs中生长抑制同源基因(Growth arrest-specific homeobox,Gax)基因的表达。结果:与正常对照组比较,胆固醇组各指标差异均有统计学意义(P<0.01);与胆固醇组比较,10.0、100.0μg·m L-1GP可逆转胆固醇所致细胞超微结构的改变,增加细胞收缩表型标志蛋白α-SMA、SM22α的表达及减少合成表型标志蛋白Epiregulin的表达(P<0.05),并能减少胆固醇所致的细胞增殖(P<0.05)。GP可逆转胆固醇所致的转录因子Gax的减少(P<0.05)。结论:GP可逆转胆固醇所致的VSMCs表型转化,并可能与增加转录因子Gax的表达有关。This study was aimed to observe the effect of gypenosides (GP) on phenotypic switching of vascular smooth muscle cells (VSMCs) induced by cholesterol. VSMCs were cultured in vitro and treated with different concentrations (1.0, 10.0, 100.0μg·mL^-1) of GP for 1 h, respectively. Then, 50.0 mg·L^-1 of cholesterol was used to process for 48 h. Morphological characteristics were observed by transmission electron microscope. Western blotting was applied to analyze the expression of smooth muscle alpha-actin (a-SMA), smooth muscle 22 alpha (SM22a) and epiregulin. Real-time PCR was applied to analyze the mRNA expression of growth arrest-specific homeobox (Gax). The results showed that compared with the control group, different indexes of the cholesterol group had statistical significance (P〈0.01). Compared with the cholesterol group, GP (10.0 and 100.0μg·mL^-1) can reverse changes of morphological characteristics induced by cholesterol, increase the expression of α-SMA, SM22a and decrease the expression of epiregulin (P〈0.05); it can also reduce the cell proliferation induced by cholesterol (P〈0.05). GP can reverse the decreasing of Gax induced by cholesterol (P〈0.05). It was concluded that GP can reverse the phenotypic switching of VSMCs induced by cholesterol. It may also be related to the increasing of Gax expression level.
