摘  要:目的:观察中药"脊髓康"对大鼠脊髓急性损伤后脊髓组织Nogo-A表达的影响。方法:选用162只SD大鼠,随机分成假手术组、模型组、强的松组、脊髓康高剂量组、脊髓康中剂量组、脊髓康低剂量组,每组27只,分别于干预后3天、7天、14天处死大鼠,以免疫组化、Western Blot及荧光定量PCR检测大鼠脊髓组织中Nogo-A的表达量。结果:脊髓损伤各组在损伤后3天时Nogo-A蛋白和m RNA表达较低,7天后迅速上升达到高峰,至14天逐渐下降。与模型组比较,脊髓康中剂量组和强的松组在损伤后3天、7天、14天时,Nogo-A蛋白和m RNA表达差异均有统计学意义(P<0.05)。结论:中药"脊髓康"能有效抑制大鼠脊髓损伤区Nogo-A表达,有利于脊髓损伤修复。This study was aimed to observe the effect of Chinese medicine di-Sui-Kang (JSK) on the expression of Nogo-A in acute spinal cord injury rats. A total of 162 SD rats were randomly divided into the sham-operated group, model group, prednisone group, JSK group of high, middle and low dose, with 27 rats in each group. The rats were sacrificed on the 3^rd, 7^th and 14^th day after intervention, respectively. The expression of Nogo-A of spinal cord tissues among all rats was detected by immunohistochemistry, western blot and fluorescence quantitative PCR. The results showed that Nogo-A protein and mRNA expression of spinal cord injury rats were decreased on the 3^rd day after injury. However, it increased rapidly to the climax on the 7^th day. And then, it gradually decreased on the 14^th day. Compared with the model group, there were statistical differences on Nogo-A protein and mRNA expression of rats in the middle dose JSK group and prednisone group on the 3^rd, 7^th and 14^th day after injury (P〈0.05). It was concluded that Chinese medicine JSK can effectively inhibit the expression of Nogo-A in acute spinal cord injury rats. It promoted the nerve regeneration in the spinal cord.
