摘  要:目的:通过观察中药安神药对与地西泮联合应用对地西泮所致小鼠学习记忆障碍的影响,探讨中药安神药对对学习记忆的保护作用及机制。方法:将具有安神作用的中药药对与地西泮联合作用于小鼠,采用Morris水迷宫进行行为学测试,并以荧光定量PCR测定小鼠海马NGFm RNA、Trk Am RNA、GABAAm RNA、TNF-αm RNA和caspase3m RNA的表达。结果:地西泮对小鼠学习记忆有损害作用,安神中药药对与地西泮联合应用,能减少小鼠Morris水迷宫测试的上平台潜伏期、游泳总路程和第1次抵原平台时间,增加穿越平台次数和目标象限时间,上调小鼠海马NGFm RNA、Trk Am RNA的表达,下调GABAAm RNA、TNF-αm RNA和caspase3m RNA的表达。结论:安神中药药对与地西泮联合应用,具有减轻地西泮所致学习记忆障碍的作用。This study was aimed to observe protective effect and mechanism of traditional Chinese medicine (TCM) tranquillization herbal pair combined with diazepam among memory impairment mice induced by diazepam. Both TCM tranquillization herbal pair and diazepam were carried out on mice. The Morris Water Maze was used for behavior proficiency test. Real time-PCR was used to detect expressions of NGFmRNA, TrkAmRNA, GABAAmRNA, TNF-αmRNA and caspase3mRNA of hippocampus tissues in mice. The results showed that diazepam had damage effect to learning and memory in mice. TCM tranquillization herbal pair combined with diazepam can reduce the latency, swimming distance and the first time to the original platform, increase the through platform number and target quadrant time in the Morris Water Maze. It upregulated the expression of NGFmRNA and TrkAmRNA, and down-regulated the expression of GABAAmRNA, TNF-amRNA and caspase3mRNA. It was concluded that TCM tranquillization herbal pair combined with diazepam can alleviate dysfunction of learning and memory caused by diazepam.
