葛根主要药效成分止泻作用研究

摘　　要：目的:测定葛根中主要药效成分的止泻作用,寻找药效物质与止泻作用相关性。方法:通过预实验考察了葛根中主要成分葛根素、大豆苷、大豆苷元的止泻作用,并选择具有明显止泻作用的葛根素和大豆苷元进行了胃排空及小肠推进率实验,结合腹泻指数,探讨两者的止泻作用,并进行了止泻量效关系研究。结果:与空白组比较,葛根素和大豆苷元均能显著增加胃内残留率(P<0.01),随给药剂量的增加胃内残留率呈现增强趋势,二者还能显著降低小肠推进率(P<0.01),随给药剂量的增加,小肠推进率呈减弱趋势;同时还能显著降低腹泻指数(P<0.01),并在一定剂量范围内存在良好量效关系。结论:葛根止泻作用的主要药效成分是葛根素、大豆苷元,为探讨葛根煨制作用机制及建立饮片质量标准参数奠定基础。This study was aimed to clarify the co-relationship between components in Radix Puerariae Lobatae and antidiarrhea] effect through determination of antidiarrheal effect related therapeutic materials. Antidiarrheal effects of puerarin, daidzin and daidzein were observed in the preliminary experiment. Gastric emptying, small intestinal propulsion rate test and diarrhea index were applied to determine the antidiarrheal effect and doseeffect relationship. The results showed that compared with the blank group, puerarin and daidzein can extremely increase the gastric remnant rate （P〈0.01）. With the dose enhancement, both of them can obviously decrease the small intestinal propulsion rate and significantly reduce the diarrhea index （P〈0.01）. It showed good dose-effect relationship within a certain range of dose. It was concluded that the main active components of antidiarrheal effect in Radix Puerariae Lobatae were puerarin and daidzein. The results laid a foundation for processing mechanism of Radix Puerariae Lobatae and established quality standard indexes.
