摘 要:目的建立UPLC测定人血浆中丙泊酚含量的方法。方法以麝香草酚为内标,用环己烷提取血浆中的丙泊酚,用UPLC测定丙泊酚含量。色谱柱为Zorbax SB-C18柱(2.1 mm×100 mm,1.8μm),流动相为乙腈-0.05%三氟乙酸水溶液(65∶35),流速0.3 ml·min-1,柱温30℃,检测波长273 nm。结果丙泊酚与血浆中其他组分分离良好,线性范围为0.1~10.0μg·ml-1,丙泊酚平均提取回收率为72.32%~81.59%,方法平均回收率为92.77%~103.16%,日内和日间精密度的RSD均小于10%。结论该方法快速、灵敏、准确,可用于临床丙泊酚血药浓度的检测和药动学研究。Objective To establish a UPLC method for determining the concentration of propofol in human plasma.Methods Thymol was used as the internal standard.After extraction with cyclohexane,the propofol concentration of all plasma samples was determined by the UPLC system with a Zorbax SB-C18column(2.1 mm × 100 mm,1.8 μm) at 30 ℃ at a flow rate of 0.3 ml·min-1and at a UV detection wavelength of 273 nm.The mobile phase was acetonitrile-0.05% TFA(65 ∶ 35).Results Propofol was separated completely from other components in the plasma.The linear range of propofol concentrations was 0.1-10.0 μg·ml-1.The extraction and recovery rate was 72.32%-81.59% and 92.77%-103.16%,respectively.Both the within-day and between-day variation coefficients were less than 10%.Conclusion This UPLC method is sensitive,quick and accurate,which can be used for monitoring propofol plasma concentrations.
