摘 要:从大豆叶片中分离到Gm Sbh1基因c DNA的完整开放阅读框(ORF)序列,对其编码的氨基酸序列、蛋白质一级、二级结构及互作蛋白等进行了生物信息学分析。结果表明:在Gm Sbh1基因c DNA序列的第156~1 293 bp处有一个完整的ORF,编码379个氨基酸;Gm Sbh1具有同源异型盒(homeoboxbox,HB)基因家族典型的KNOX1、KNOX2、ELK和Homeodomain(HD)结构域;检索到18条与Gm Sbh1核酸序列一致性大于80%的序列;Gm SBH1为亲水性蛋白,不具有信号肽,分子量为42.37 k Da,其中丝氨酸(Ser)、天冬氨酸(Asn)和亮氨酸(Leu)分别为占10%、9.8%和8.4%,可能发生磷酸化的位点分别在Ser(13个)、酪氨酸(Tyr)(2个)和苏氨酸(Thr)(1个)上;二级结构预测结果显示,Gm SBH1序列存在α-螺旋(占39.31%)、β-转角(占5.28%)、延伸链(占8.44%)和无规则卷曲(占46.97%),不存在跨膜螺旋;蛋白互作预测表明,Gm SBH1与拟南芥中的RPL、BEL1、AT4G32980.1-P、BLH2和BLH3等蛋白的互作程度较高,其中与BEL1共表达的几率较高。The cDNA sequences of GmSbhl gene in soybean were isolated, and its bioinformatics character was analyzed. The results showed that there was an intact ORF in 156-1 293 bp , and it encoded a 42. 37 kDa protein with 379 amino acids. The GmSBH1 protein contains the KNOX1, KNOX2, ELK and Homeodomain (HD). There were 18 homologous sequences that have more than 80% identity with GmSbhl. GmSBH1 was a hydrophilic protein without signal peptide and contained 10% Set, 9. 8% Asn and 8.4% I.eu, the phosphorylation sites were located at Set( 13), Tyr(2) and Thr( 1 ), respectively. Sec- ondary structure prediction results showed that the GmSBttl protein contains alpha-helix (39. 31% ), beta turn (5.28%), extended strand(8.44% ) and random coil (46.97%) , but without transmembrane helical structure. The probability of Gm- SH1 interacted with RPL, BELl, AT4G32980. l-P, BLH2 and BLH3 was higher, and coexpression with BELl in Arabibidop-sis.
