摘  要:目的:探讨姜黄素对老年性痴呆(Alzhelmer's disease,AD)大鼠海马区β-淀粉样前体蛋白(β-amyloid protein precursor,β-APP)和β-分泌酶(amyloidβsecretase1,BACE1)基因表达的影响。方法:采用Aβ1-40右侧海马区注射,制备AD大鼠模型,腹腔注射姜黄素给予治疗,采用ELISA法检测血清与海马组织中Aβ的含量,免疫组织化学法检测海马区神经元中β-APP蛋白表达,RT-PCR法检测海马组织中β-APP和BACE1 mRNA表达。结果:造模后大鼠血清和脑组织海马中Aβ含量显著升高,脑组织海马区神经元中β-APP蛋白表达、β-APP mRNA和BACE1 mRNA表达明显增加。与模型组比较,姜黄素高剂量组、低剂量组β-APP蛋白表达、β-APP mRNA和BACE1 mRNA明显降低,差异均有统计学意义(P<0.05)。结论:姜黄素可通过对BACE1 mRNA调控作用,阻断β-APP裂解进程,抑制Aβ在脑内生成与沉积,从而减少Aβ对神经元的毒性作用。Objective:To explore the effect of genetic expression of hippocampus' β-APP and BACE1 by curcumin. Methods:Injecting the right hippocampus by Aβ-40, preparing a model of AD rats,intraperitoneal injection of curcumin, using ELISA to detect the content of Aβ in serum and hippocampus,immune histochemical method to detect the hippocampus neuron in β-APP protein expression,rt-pcr method to detect beta APP and BACElmRNA expression in hippocampus. Results:Aβ had been remarkable rise, β-APP expression of protein, β-APPmRNA and BACElmRNA expression remarkable increase after the molding. Comparing the model group, Curcumin in high dose group,low dose group of beta APP protein expression, beta APPmRNA and significantly lower BACE1 mRNA, difference had statistical significance ( P 〈 0.05 ). Conclusion : Curcumin can interdict β-APP splitting decomposition, Inhibition of Aβfonnation and deposition in the brain by regulating BACE1 mRNA,therefore reduce the toxic effect of nerve cell.
